| Abstrak (Bhs. Indonesia) | Kitin merupakan polimer yang tersusun atas β-1,4- N-asetil-D-glukosamin (GlcNac) dengan kelimpahan terbanyak kedua setelah selulosa. Sumber kitin yang banyak dapat menjadi limbah dan harus diurai kembali menjadi komponennya melalui biodegradasi secara enzimatis menggunakan bakteri penghasil kitinase. Laboratorium Mikrobiologi Fakultas Biologi memiliki 30 isolat bakteri yang diisolasi dari sedimen mangrove Pantai Logending, Kebumen yang belum diketahui aktivitas kitinasenya. Aktivitas kitinase dapat dipengaruhi oleh faktor pH dan suhu. Tujuan penelitian ini yaitu untuk mengetahui aktivitas kitinase bakteri kitinolitik terbaik asal sedimen mangrove Pantai Logending Kebumen pada pH dan suhu yang berbeda, serta mengetahui identitasnya. Penelitian menggunakan Rancangan Acak Lengkap (RAL) dengan analisis menggunakan One Way ANOVA dan uji lanjut Tukey HSD. Variabel bebas berupa pH yaitu variasi nilai pH 4, 5, 6, 7, dan 8, dan suhu yaitu variasi 30 oC, 35 oC, 40oC, 45oC, dan 50oC. Masing-masing perlakuan dilakukan pengulangan sebanyak 4 kali. Variabel terikat yaitu aktivitas kitinase. Uji kemampuan kitinolitik isolat bakteri menggunakan medium kitin agar dan uji aktivitas kitinase menggunakan metode DNS. Identifikasi isolat bakteri merujuk pada buku Bergey’s Manual of Systematic Bacteriology. Hasil penelitian menunjukkan bahwa sebanyak 22 isolat dari 30 isolat bakteri memiliki aktivitas kitinolitik. Isolat bakteri LG74 memiliki kemampuan paling tinggi dengan Indeks Kitinolitik sebesar 0,325. Aktivitas kitinase isolat LG74 memiliki kondisi optimum pada pH 6 dan suhu 45 oC dengan nilai aktivitas secara berturut-turut adalah 0,3068 U/mL dan 0,6847 U/mL. Hasil identifikasi isolat bakteri LG74 termasuk spesies anggota dari Genus Bacillus. |
| Abtrak (Bhs. Inggris) | Chitin is a polymer composed of β-1,4-N-acetyl-D-glucosamine (GlcNac) which is widely found in nature. The abundance of chitin is the second largest after cellulose. A great abundance of chitin sources must be degraded into its components through enzymatic biodegradation using chitinase-producing bacteria. The Microbiology Laboratory of the Faculty of Biology has 30 bacterial isolates isolated from mangrove sediments at Logending Beach, Kebumen whose chitinase activity is not yet known. Chitinase activity can be influenced by pH and temperature factors. This study aimed to determine the best chitinolytic bacterial isolate's chitinase activity at different pH and temperatures and its identity which has been isolated from mangrove sediments at Logending Kebumen Beach. The study was conducted experimentally using a Completely Randomized Design (CRD) which was analysed with One Way ANOVA and Tukey HSD further test. The independent variables used were pH there are 4, 5, 6, 7, and 8, and temperature there are 30 oC, 35 oC, 40 oC, 45 oC, and 50 oC, each treatment were repeated 4 times. The dependent variable was chitinase activity. The chitinolytic ability test of bacterial isolates was carried out using a chitin agar medium and the chitinase activity test was carried out using the DNS method. Identification of bacterial isolates refers to Bergey's Manual of Systematic Bacteriology. The results showed that 22 isolates from 30 bacterial isolates had chitinolytic activity. The bacterial isolate LG74 had the highest ability with a Chitinolytic Index of 0,325 U/mL. The chitinase activity of LG74 isolate had optimum conditions at pH 6 and a temperature of 45 oC with activity values of 0,3068 U/mL and 0,6847 U/mL, respectively. The results of identifying the bacterial isolate LG74 include species belonging to the Genus Bacillus. |
