Indonesia) | ABSTRAK POTENSI YOGURT KECAMBAH KACANG TOLO TERHADAP PERUBAHAN KADAR TUMOR NECROSIS FACTOR-ALPHA (TNF-α) PLASMA PENDERITA DIABETES MELITUS TIPE II Windi Sri Lestari Handayani1 , Hery Winarsi2 , Teguh Jati Prasetyo2 Latar Belakang: Hiperglikemia pada DM menimbulkan peningkatan produksi ROS. Ketidakseimbangan antara peningkatan ROS dengan ketersediaan antioksidan dapat menimbulkan kondisi stres oksidatif yang berdampak pada kerusakan jaringan dan penurunan imunitas tubuh. Kedua kondisi tersebut dapat meningkatkan resiko terjadinya inflamasi dan meningkatkan produksi sitokin pro-inflamasi TNF-α. Kadar TNF-α yang tinggi dan dapat menyebabkan komplikasi makro maupun mikrovaskuler. Produk pangan kaya antioksidan seperti yogurt kecambah kacang tolo (yocamcalo) diharapkan mampu memperbaiki status imun dan menurunkan kadar TNF-α plasma penderita DM Tipe II. Metodologi: Penelitian eksperimental ini menggunakan rancangan Randomized Controlled Trial (RCT) single blind. Penelitian dilaksanakan pada Februari-Maret 2022 di Laboratorium Pangan dan Gizi Unsoed dan Desa Purwosari, Kabupaten Banyumas. Responden berjumlah 18 orang dibagi menjadi kelompok yocamcalo dan kelompok plasebo. Kadar TNF-α plasma diukur sebelum dan setelah intervensi menggunakan ELISA reader. Data karakteristik responden dianalisis menggunakan uji Independent Sample T Test dan Mann Whitney, penurunan kadar TNF-α plasma dianalisis menggunakan uji Wilcoxon. Hasil Penelitian: Setelah dua bulan pemberian Yocamcalo 200 mL/hari, terjadi penurunan rata-rata kadar TNF-α plasma dari 54,58 pg/mL menjadi 34,47 pg/ mL (p=0,015). Kesimpulan: Pemberian Yocamcalo sebanyak 200mL/hari selama dua bulan dapat memberikan efek berupa penurunan kadar TNF-α plasma pada penderita DM Tipe II. Inggris) | ABSTRACT POTENTIAL OF TOLO BEAN SPROUT YOGURT ON CHANGE TUMOR NECROSIS FACTOR-ALPHA (TNF-α) PLASMA PATIENTS WITH TYPE II DIABETES MELLITUS Windi Sri Lestari Handayani1 , Hery Winarsi2 , Teguh Jati Prasetyo2 Background: Hyperglycemia in DM causes an increase in ROS production. The imbalance between the increase in ROS and the availability of antioxidants can cause oxidative stress conditions that have an impact on tissue damage and decreased immunity. Both conditions can increase the risk of inflammation and increase the production of the pro-inflammatory cytokine TNF-α. TNF-α levels are high and can cause macro and microvascular complications. Antioxidant-rich food products such as tolo bean sprout yogurt (yocamcalo) are expected to improve immune status and reduce plasma TNF-α levels in patients with Type II DM. Methodology: This experimental study used a single blind Randomized Controlled Trial (RCT). The research was carried out in February-March 2022 at the Food and Nutrition Laboratory of Unsoed and Purwosari Village, Banyumas Regency. The 18 respondents were divided into the yocamcalo group and the placebo group. Plasma TNF-α levels were measured before and after the intervention using an ELISA reader. The respondent's characteristic data were analyzed using the Independent Sample T Test and Mann Whitney, the decrease in plasma TNF- levels was analyzed using the Wilcoxon test. Results: After two months of administration of Yocamcalo 200 mL/day, the average plasma TNF- level decreased from 54.58 pg/mL to 34.47 pg/mL (p=0.015). Conclusion: Giving Yocamcalo as much as 200mL/day for two months can have an effect in the form of decreasing plasma TNF- levels in patients with Type II DM.
