| Abstrak (Bhs. Indonesia) | Indonesia memiliki sumber daya alam yang melimpah dan beragam diantaranya berpotensi dalam pengobatan, salah satunya jamur lingzhi (Ganoderma lucidum). Jamur ini mengandung komponen bioaktif seperti terpenoid, polisakarida, nukleotida, sterol, steroid, asam lemak dan protein/peptida yang memiliki potensi dalam pengobatan. G. lucidum sudah banyak dibudidayakan di Indonesia dan terbukti memiliki efek farmakologis seperti antidiabetes, antitumor, antimikroba, dan antiinflamasi. Terpenoid merupakan senyawa utama pada ekstrak etanol G. lucidum yang dapat berperan sebagai antiinflamasi. Senyawa bioaktif G. lucidum sebagai antiinflamasi terkait dengan penghambatan aktivitas NF-κB yang merupakan faktor transkripsi pengatur ekspresi gen proinflamasi selama inflamasi. Aktivitas NF-κB perlu ditekan karena dapat memperparah reaksi inflamasi. Pelepasan mediator inflamasi yang tidak terkontrol berdampak negatif pada ginjal sehingga dapat menyebabkan disfungsi ginjal yang ditandai dengan meningkatnya kadar ureum dan kreatinin di dalam darah. Hal ini tentunya akan berdampak negatif bagi kesehatan tubuh. Penelitian ini bertujuan untuk mengetahui pengaruh serta dosis efektif ekstrak G. lucidum terhadap penurunan kadar ureum dan kreatinin darah tikus putih jantan galur Wistar model inflamasi terinduksi Complete Freund’s Adjuvant (CFA). Penelitian ini merupakan penelitian eksperimental menggunakan Rancangan Acak Lengkap (RAL). Tikus dibagi menjadi enam kelompok. Kelompok Kontrol Sehat (KS) diberi aquades. Kelompok Kontrol Negatif (KN), Kontrol Positif (KP) dan Kelompok Perlakuan (P1, P2, dan P3) diinduksi CFA 0,1 ml sebagai penginduksi inflamasi. Kelompok Perlakuan (P1, P2, dan P3) diberi perlakuan ekstrak jamur G. lucidum dengan variasi dosis 250, 500, 750 mg.kg-1 BB. Variabel bebas berupa dosis ekstrak G. lucidum 250, 500, 750 mg.kg-1 BB dan variabel terikat berupa kadar ureum dan kreatinin. Parameter yang diamati kadar ureum dan kreatinin sebagai penanda fungsi ginjal. Pada hari ke 21 dilakukan pengambilan darah tikus terinduksi CFA yang diberi ekstrak G. lucidum untuk mengukur kadar ureum dan kreatinin. Data hasil pengukuran kadar ureum dan kreatinin dianalisis dengan uji ANOVA kemudian dilanjutkan dengan uji Duncan. Hasil penelitian menujukkan bahwa pemberian ekstrak etanol G. lucidum mampu menurunkan kadar kreatinin dan ureum tikus model inflamasi yang diinduksi CFA. Ekstrak etanol G. lucidum dengan dosis 750 mg.kg-1 BB dapat menurunkan kadar ureum dan kreatinin tikus dengan rata-rata kadar kreatinin sebesar 0,75±0,32 mg.dL-1 dan ureum sebesar 57,50±4,82 mg.dL-1 dengan penurunan kadar kreatinin dan ureum yang dibandingkan kontrol negatif masing-masing sebesar 60,93% dan 20,16%. Kesimpulan dari penelitian ini adalah ekstrak etanol G. lucidum dengan dosis 750 mg.kg-1 BB efektif dalam menurunkan kadar ureum dan kreatinin tikus model inflamasi terinduksi CFA. |
| Abtrak (Bhs. Inggris) | Indonesia has abundant natural resources and various of them have potential in medicine, one of which is the lingzhi mushroom (Ganoderma lucidum). This mushroom contains bioactive components such as terpenoids, polysaccharides, nucleotides, sterols, steroids, fatty acids and proteins/peptides that have medicinal potential. G. lucidum has been widely cultivated in Indonesia and shown pharmacological effects such as antidiabetic, antitumor, antimicrobial, and anti-inflammatory. Terpenoids are the main compounds in the ethanolic extract of G. lucidum which can act as anti-inflammatory. The bioactive compound G. lucidum as an anti-inflammatory is associated with suppression of the expression of iNOS, COX-2, and NF-κB which are regulators of proinflammatory gene expression during inflammation. The expression of the enzyme needs to be supressed because they can exacerbate the inflammatory reaction. Uncontrolled release of inflammatory mediators has a negative impact on the kidneys so that it can cause kidney dysfunction. Increased levels of urea and creatinine is one of the biomarkers of kidney dysfunction, so it is necessary to check the levels of urea and creatinine. This study aimed to determine the effect and effective dose of G. lucidum extract on reducing blood urea and creatinine levels of white male rat (Rattus norvegicus) Wistar strain which was induced by Complete Freund's Adjuvant (CFA). This research is an experimental study using Completely Randomized Design (CRD). Rats were divided into six groups. The Healthy Control Group (KS) was given aquades. Negative Control Group (KN), Positive Control (KP) and Treatment Group (P1, P2, and P3) were induced with 0.1 ml CFA as an inflammation inducer. The treatment groups (P1, P2, and P3) were treated with G. lucidum mushroom extract with various doses of 250, 500, 750 mg.kg-1 BW. The independent variable was the dose of G. lucidum extract 250, 500, 750 mg.kg-1 BW and the dependent variable was urea and creatinine levels. Parameters observed were urea and creatinine levels as markers of kidney function. On day 21, blood samples from CFA-induced rats were taken with G. lucidum extract to measure urea and creatinine levels. The measurement data of urea and creatinine levels were analyzed by ANOVA test, and it would be continued with Duncan's test. The results showed that the administration of the ethanolic extract of G. lucidum was able to reduce creatinine and urea levels in rats induced by CFA-induced inflammation. Ethanol extract of G. lucidum with a dose of 750 mg.kg-1 BW can reduce urea and creatinine levels in rats with an average creatinine level of 0.75±0.32 mg.dL-1 and urea by 57.50±4.82. mg.dL-1 with a decrease in creatinine and urea levels compared to negative controls by 60.93% and 20.16%, respectively. The conclusion of this study is that the ethanol extract of G. lucidum at a dose of 750 mg.kg-1 BW was effective in reducing urea and creatinine levels in rats modeled by CFA-induced inflammation. |
