| Abstrak (Bhs. Indonesia) | Bahan organik di area mangrove terbentuk karena adanya sedimen yang bercampur dengan serasah daun mangrove. Dekomposisi bahan organik dengan kandungan selulosa yang tinggi pada lingkungan mangrove mengakibatkan adanya produksi enzim ekstraseluler oleh mikroorganisme. Mikroorganisme di lingkungan mangrove berperan sebagai sumber bakteri selulolitik dapat ditemukan pada kayu lapuk, lumpur, tanah dan serasah daun mangrove. Hidrolisis selulosa menjadi senyawa yang lebih sederhana pada mangrove melibatkan lebih dari satu jenis enzim. Penelitian ini bertujuan untuk mengetahui kemampuan isolat dalam mendegradasi selulase, mengetahui suhu dan pH optimal aktivitas enzim selulase, dan mengetahui identitas isolat bakteri terseleksi asal sedimen mangrove pantai Logending Kebumen Jawa Tengah. Penelitian dilakukan dengan metode survei, dan data hasil penelitian dianalisis secara deskriptif. Penelitian dilakukan selama 5 bulan di Laboratorium Mikrobiologi Fakultas Biologi. Tahapan penelitian meliputi screening kemampuan isolat bakteri dalam mendegradasi selulase pada medium Carboxymethylcellulose (CMC), penentuan suhu dan pH optimal aktivitas enzim selulase dengan metode Dinitrosalicylic Acid (DNS) pada variasi suhu 30°C, 35°C; 40°C; 45°C dan variasi pH 4; 4,5; 5; 6 dan mengkarakterisasi isolat bakteri terseleksi dengan metode Manual. Identifikasi bakteri mengacu pada buku “Bergey’s Of Determinative Bacteriology”. Hasil penelitian diperoleh indeks selulolitik tertinggi dari 10 isolat asal sedimen mangrove ditunjukkan oleh isolat LG21 yakni 1,25. pH optimal aktivitas selulolitik isolat asal sedimen mangrove pantai Logending pH 6 yakni 1,952 U/mL dan suhu optimal 45ºC yakni 1,93 U/mL. Jumlah total biomassa bakteri selulolitik setelah waktu produksi 5,74x1014 CFU/mL. Identitas isolat bakteri selulolitik asal sedimen mangrove pantai Logending dengan kode isolat LG21 termasuk spesies anggota genus Bacillus. |
| Abtrak (Bhs. Inggris) | Organic matter in the mangrove area is formed due to the presence of sediment mixed with mangrove leaf litter. Decomposition of organic matter with high cellulose content in the mangrove environment results in the production of extracellular enzymes by microorganisms. Microorganisms in the mangrove environment act as a source of cellulolytic bacteria that can be found in weathered wood, mud, soil and mangrove leaf litter. Hydrolysis of cellulose into simpler compounds in mangroves involves more than one type of enzyme. This study aims to determine the ability of isolates to degrade cellulase, determine the optimal temperature and pH of cellulase enzyme activity, and determine the identity of selected bacterial isolates from mangrove sediments at the Logending beach, Kebumen, Central Java. The research was conducted using a survey method, and the research data were analyzed descriptively. The research was conducted for 5 months at the Microbiology Laboratory, Faculty of Biology. The research stages included screening the ability of bacterial isolates to degrade cellulase on Carboxymethylcellulose (CMC) medium, determining the optimal temperature and pH for cellulase enzyme activity using the Dinitrosalicylic Acid (DNS) method at various temperatures of 30°C, 35°C; 40°C; 45°C and variation of pH 4; 4.5; 5; 6 and characterizing the selected bacterial isolates using the Manual method. Identification of bacteria refers to the book "Bergey's Of Determinative Bacteriology". The results showed that the highest cellulolytic index of 10 isolates from mangrove sediments was indicated by isolate LG21, which was 1.25. The optimal pH for cellulolytic activity of isolates from mangrove sediments at the Logending beach is pH 6, which is 1.952 U/mL and the optimal temperature is 45ºC, which is 1.93 U/mL. The total amount of cellulolytic bacterial biomass after production time was 5.74x1014 CFU/mL. Identity of isolates of cellulolytic bacteria from mangrove sediments at the Logending beach with isolate code LG21 including species belonging to the genus Bacillus. |
