| Abstrak (Bhs. Indonesia) | Latar Belakang: Sleep deprivation (SD) masih menjadi suatu masalah kesehatan yang dikaitkan dengan peningkatan morbiditas dan mortalitas. Sleep deprivation melalui mekanisme sistem saraf simpatis dan aksis HPA (Hypothalamic-Pituitary-Adrenal) dapat menginduksi nuclear factor (NF)-B sehingga meningkatkan transkripsi sitokin pro-inflamasi, salah satunya IFN-. IFN- terlibat dalam regulasi inflamasi dan apoptosis. Dampak dari sleep deprivation dapat dikoreksi dengan sleep recovery (SR) dengan mengembalikan aktivitas antioksidan dalam menghambat stres oksidatif. Penelitian mengenai sitokin IFN- terkait perbedaan kadarnya setelah induksi sleep deprivation pada model hewan coba belum banyak dilakukan. Tujuan: Mengetahui perbedaan kadar Interferon Gamma (IFN-) pada tikus putih (Rattus norvegicus) jantan pasca induksi berbagai model stres sleep deprivation. Metode: Penelitian ini merupakan penelitian eksperimental dengan desain post-test only with control group. Penelitian ini menggunakan 25 ekor tikus putih (Rattus norvegicus) jantan galur Wistar yang dibagi secara acak menjadi 5 kelompok percobaan yaitu kontrol, PSD (20 jam SD/hari selama 5 hari), TSD (24 jam SD/hari selama 5 hari), PSD+SR (20 jam SD/hari selama 5 hari dilanjutkan SR selama 5 hari), dan TSD+SR (24 jam SD/hari selama 5 hari dilanjutkan SR selama 5 hari). Hasil: Uji One-way ANOVA menunjukkan tidak terdapat perbedaan yang signifikan kadar IFN- antar kelompok hewan coba (p=0,283). Rerata kadar IFN- tertinggi didapatkan pada kelompok TSD (1167,39±423,52 pg/ml). Peningkatan kadar IFN- tertinggi terdapat pada kelompok TSD yakni 1,12-fold. Kesimpulan: Tidak terdapat perbedaan signifikan kadar sitokin pro-inflamasi IFN- pada tikus putih (Rattus norvegicus) jantan pasca induksi berbagai model stres sleep deprivation. |
| Abtrak (Bhs. Inggris) | Introduction: Sleep deprivation (SD) still considered as a health problem that is associated with increased morbidity and mortality. Sleep deprivation through the mechanism of sympathetic nervous system and the HPA (Hypothalamic-Pituitary-Adrenal) axis can induce nuclear factor (NF)-B thereby increasing pro-inflammatory cytokines transcription, including IFN-. IFN- is involved in the regulation of inflammation and apoptosis. The impact of sleep deprivation can be corrected by sleep recovery (SR) by restoring antioxidant activity and inhibiting oxidative stress. Study on IFN- related to sleep deprivation in animal models has not been widely carried out. Objective: To determine the differences of Interferon Gamma (IFN-) levels in male albino rats (Rattus norvegicus) after induction various stress models of sleep deprivattion. Methods: True experimental study with postest only and control group design. Twenty five male albino rats were distributed into 5 groups consist of control group, PSD (20 hours of SD/day for 5 days), TSD (24 hours of SD/day for 5 days), PSD+SR (20 hours SD/day for 5 days then SR for 5 days), dan TSD+SR (24 hours SD/day for 5 day then SR for 5 day). Measurement of IFN- using Milliplex Bead-based Multiplex Assays. Results: One-way ANOVA test showed no significant difference in IFN- levels (p=0.283). The highest mean IFN- level was in the TSD group (1167.39±423.52 pg/ml). The increase in IFN- level was highest in the TSD group (1.12 fold). Conclusions: There is no significant difference of the IFN- level in male albino rats (Rattus norvegicus) after induction various stress models of sleep deprivation. |
