| Abstrak (Bhs. Indonesia) | Oral Squamous Cell Carcinoma (OSCC) merupakan kanker dengan prevalensi kasus yang tinggi di Indonesia dengan prognosis buruk mencapai 50% dari total. mRNA THBS2 diketahui memiliki peran terhadap tingkat keganasan sehingga potensial untuk menjadi kandidat biomarker OSCC agar dapat dideteksi sejak dini. Penelitian ini bertujuan untuk mengetahui perbedaan ekspresi mRNA THBS2 dan jumlah platelet, serta hubungan perubahan ekspresi mRNA THBS2 dengan jumlah platelet pada low dan high grade OSCC. Penelitian observasional analitik dengan rancangan cross sectional menggunakan jaringan FFPE sebanyak 18 sampel OSCC yang dipilih random. Metode penelitian melalui tahapan deparafinasi, ekstraksi RNA, dan sintesis cDNA. Quantifikasi ekspresi menggunakan qPCR dan data jumlah platelet dikoleksi dari rekam medis pasien OSCC. Analisis statistik menggunakan software GenEX MultiD dan SPSS dengan uji Independent t-test dan Pearson. Analisis in silico menggunakan website STITCH dan STRING. Hasil analisis menunjukkan ekspresi mRNA THBS2 dengan fold change sebesar 1,3371 (p>0,05) pada kelompok high grade, tidak terdapat perbedaan pada jumlah platelet, serta terdapat hubungan antara perubahan ekspresi gen dengan jumlah platelet pada low grade dan high grade. Analisis in silico menunjukkan peran dari THBS2 dalam meregulasi CD36 pada pertumbuhan sel kanker. Simpulan dari penelitian ini menunjukan peran THBS2 pada pertumbuhan sel kanker. |

| Abtrak (Bhs. Inggris) | Oral Squamous Cell Carcinoma (OSCC) is a cancer with a high prevalence of cases in Indonesia with a poor prognosis reaching 50% of the total. THBS2 mRNA is known to have a role in the level of malignancy so that it has the potential to be a candidate for OSCC biomarkers so that it can be detected early. This study aims to determine differences in THBS2 mRNA expression and platelet count, as well as the relationship between changes in THBS2 mRNA expression and platelet count in low and high grade OSCC. An analytical observational study with a cross sectional design using the FFPE network as many as 18 samples of OSCC were selected randomly. The research method is through the stages of deparaffination, RNA extraction, and cDNA synthesis. Expression quantification using qPCR and platelet count data were collected from medical records of OSCC patients. Statistical analysis using software GenEX MultiD and SPSS with Independent t-test and Pearson test. In silico analysis using STITCH and STRING websites. The results of the analysis showed that the THBS2 mRNA expression with a fold change of 1.3371 (p>0.05) in the high grade group, there was no difference in the number of platelets, and there was a relationship between changes in gene expression and the number of platelets in the low grade and high grade. In silico analysis demonstrated the role of THBS2 in regulating CD36 in cancer cell growth. The conclusion of this study shows the role of THBS2 on cancer cell growth. |
