| Abstrak (Bhs. Indonesia) | Penelitian kali ini bertujuan untuk melakukan seleksi terhadap 9 isolat bakteri endofit akar padi (KR4, KR5, KR7, SR1, SR5, SR7, SB1, SB3, SM1) dengan jamur R solani secara in vitro, mengetahui kurva pertumbuhan, produksi dan aktivitas enzim kitinase bakteri endofit hasil seleksi. Penelitian dilaksanakan di Laboratorium Perlindungan Tanaman Fakultas Pertanian dan Laboratorium Riset, Universitas Jenderal Soedirman, Purwokerto. Penelitian dimulai pada bulan Desember 2019 hingga Agustus 2020. Rancangan yang digunakan yaitu Rancangan Acak Lengkap (RAL) dengan 10 perlakuan dan 3 ulangan.Variabel yang diamati adalah kemampuan penghambatan bakteri endofit terhadap patogen R. solani, kurva pertumbuhan, produksi serta aktivitas enzim kitinase bakteri endofit dan kemampuan penghambatan enzim kitinase bakteri endofit terhadap patogen R. solani. Hasil penelitian menunjukan bahwa hasil seleksi pada uji antagonis menggunakan koloni, isolat KR7 dan SB3 merupakan dua isolat terbaik dalam menghambat jamur R. solani . Pertumbuhan bakteri KR7 dan SB3 mencapai fase stasioner pada waktu inkubasi 12 jam. Produksi dan aktivitas enzim kitinase isolat KR7 tertinggi diketahui pada waktu inkubasi 24 jam sebesar 0,0944 U/mL. Begitu juga dengan isolat SB3, produksi dan aktivitas enzim tertinggi diketahui pada waktu inkubasi 24 jam sebesar 0,0916 U/mL. |

Inggris) | This study was aimed to select 9 isolates of rice root endophytic bacteria (KR4, KR5, KR7, SR1, SR5, SR7, SB1, SB3, SM1) to control R. solani in vitro, to know the growth curve of the selected endophytic bacteria and to know the production curve along with the activity of the chitinase enzyme of the selected endophytic bacteria. The research was carried out at the Plant Protection Laboratory and Research Laboratory, Faculty of Agriculture, Jenderal Soedirman University, Purwokerto. The research was conducted from December 2019 until August 2020. The design used was a completely randomized design (CRD) with 10 treatments and 3 replications. The variables observed were the inhibition ability of endophytic bacteria against R. solani, growth curves, production, and activity of the bacterial chitinase enzyme endophytic and the ability of endophytic bacteria chitinase enzyme inhibition against R. solani. The results showed that the isolate of KR7 and SB3 were two best isolates in inhibiting R.solani based on the antagonist assay. The growth of KR7 and SB3 bacteria reached a stationary phase at an incubation time of 12 hours. The highest production and activity of chitinase enzyme KR7 isolate was known at the 24-hour incubation time of 0,944 U/mL. As well as the SB3 isolate, the highest production and activity of chitinase enzyme was known at the 24-hour incubation time of 0,0916 U/mL. |
