Abstrak (Bhs. Indonesia)Ekosistem mangrove Logending terdiri dari berbagai organisme yang termasuk dalam kelompok sistemik yang berbeda seperti tumbuhan, bakteri, jamur, mikroalga, invertebrata, burung, dan mamalia. Aspek biologi hutan mangrove dipengaruhi oleh serapan mineral oleh tumbuhan, aktivitas biologi seluruh biota di hutan mangrove, dekompposisi, produksi serasah, dan pelapukan batang mangrove yang mati. Serasah dan pelapukan batang mangrove memungkinkan adanya kandungan pektin yang tinggi di dalam tanah, juga dimungkinkan banyak ditemukan mikroorganisme yang mampu mendegradasi pektin di dalam tanah. Bakteri yang mampu mendegradasi pektin disebut bakteri pektinolitik. Bakteri pektinolitik mampu mendegradasi pektin dengan menghasilkan enzim pektinase. Pektinase adalah sekelompok enzim yang mengkatalisis polimer pektin dengan reaksi degradasi melalui de-polimerisasi dan de-esterifikasi. Pektinase ini memiliki aplikasi yang luas dalam industri makanan, tekstil, dan kertas.
Tujuan penelitian ini adalah untuk mengetahui kemampuan isolat bakteri ekosistem mangrove Logending dalam mendegradasi pektin, mengetahui aktivitas pektinasenya, dan mengetahui identitas bakteri tersebut. Metode penelitian yang digunakan adalah metode survei dengan parameter utama yang diamati adalah indeks hidrolisis pektinolitik dan aktivitas pektinase. Skrining bakteri pendegradasi pektin diamati secara kualitatif pada media agar Vincent. Uji aktivitas pektinase dilakukan secara kuantitatif dengan menggunakan metode DNS. Identifikasi bakteri potensial mengacu pada Bergey's Manual of Systematic Bacteriology.
Hasil penelitian diperoleh 28 isolat bakteri yang mampu mendegradasi pektin. Tiga isolat potensial LG13, LG101, dan LG135 mampu mendegradasi pektin tertinggi yang memiliki indeks hidrolisis pektinolitik berturut-turut yaitu 3,67, 3,48, dan 3,38 serta aktivitas pektinase masing-masing sebesar 166,6 U / mL, 111 U / mL, dan 92,2 U / mL. Ketiga isolat tersebut diidentifikasi sebagai anggota spesies dari genus Bacillus.

Inggris)Logending mangrove ecosystem is composed of a wide range of organisms belonging to different systemic groups including plants, bacteria, fungi, microalgae, invertebrates, birds, and mammals. Biological aspects of mangrove forests are influenced by mineral uptake by plants, biological activities of all biota in mangrove forests, decomposition, litter production, and weathering of dead mangrove stems. Litter and weathering of mangrove stems are possible in the presence of high pectin content in the soil, it is also possible to find many microorganisms that are able to degrade pectin in the soil. The bacteria that are capable to degrade pectin are called pectinolytic bacteria. Pectinolytic bacteria are capable of degrading pectin by producing the pectinase enzyme. Pectinases are a group of enzymes that catalyze the polymer of pectin by a degradation reaction through de-polymerization and de-esterification. These pectinases have wide applications in the food, textile, and paper industry.
The objectives of this research were to know the capability of bacterial isolates of Logending mangrove ecosystem in degrading pectin, to know their pectinase activity, and to know the identity of bacteria. The research method used was the survey method with the main parameters observed were pectinolytic hydrolysis index and pectinase activity. Screening for pectin degrading bacteria was observed qualitatively on Vincent's agar medium. Pectinase activity assay was carried out quantitatively by using the DNS method. The identification of potential bacteria was referred to Bergey’s Manual of Systematic Bacteriology.
The results obtained 28 bacterial isolates that were able to degrade pectin. The three highest potential isolates LG13, LG101, and LG135 were capable of degrading pectin which had a pectinolytic hydrolysis index of 3.67, 3.48, and 3.38 respectively, and their pectinase activity of 166.6 U/mL, 111 U/mL, and 92.2 U/mL respectively. The three isolates were identified as species members of the genus Bacillus.
