| Abstrak (Bhs. Indonesia) | Anggrek Hibrid Vanda ‘Propathom gold’ x Vanda ‘Robert delight blue’ merupakan hasil persilangan vanda yang mudah berbunga dengan bunga berukuran besar dan memilki wana yang indah. Melalui persilangan dengan usaha perbaikan genetik diharapkan dapat meningkatkan kualitas vanda. Untuk mengetahui keindahan bunga hasil persilangan interspesifik tersebut diperlukan teknik kultur in vitro untuk meningkatkan kuantitas. Penelitian ini dilakukan dengan tujuan untuk mengetahui pengaruh antara IBA dan BAP terhadap pertumbuhan plantlet Anggrek Vanda ‘Propathom gold’ x Vanda ‘Robert delight blue’ dan menentukan interaksi IBA dan BAP yang paling baik untuk pertumbuhan plantlet Anggrek Vanda ‘Propathom gold’ x Vanda ‘Robert delight blue’. Penelitian dilakukan secara eksperimental dengan rancangan dasar Rancangan Acak Lengkap (RAL) dengan pola perlakuan faktorial. Faktor pertama yaitu konsentrasi IBA (I) dengan 4 taraf yaitu I1 : 0 ppm, I2 : 0,5 ppm, I3 : 1 ppm, dan I4 : 1,5 ppm. Faktor kedua yaitu konsentrasi BAP (B) dengan 4 taraf yaitu B1 :0 ppm, B2 : 0,5 ppm, B3 : 1 ppm dan B4 : 1,5 ppm. Setiap kombinasi perlakuan diulang sebanyak 3 kali, sehingga terdapat 48 unit percobaan.Variabel yang diamati adalah pertumbuhan plantlet, dengan parameter yang diukur meliputi jumlah daun, jumlah akar, dan tinggi plantlet. Data yang diperoleh dianalisis dengan Analisis Ragam dengan tingkat kepercayaan 95%, dan perlakuan yang berpengaruh nyata, dilanjutkan dengan uji Duncan Multiple Range Test (DMRT) pada tingkat kepercayaan 95%. Hasil penelitian menunjukkan interaksi yang terbaik untuk pertumbuhan akar yaitu IBA 1 ppm dan BAP 1,5 ppm sebanyak 2,11 buah, serta untuk pertambahan tinggi plantlet yaitu IBA 1,5 ppm dan BAP 1 ppm hingga 0,98 cm. Pemberian secara mandiri IBA 1 ppm terbaik dalam memacu pertumbuhan akar sebanyak 1,88 buah, dan IBA 1,5 ppm terbaik dalam meningkatkan tinggi plantlet hingga 0,91 cm. Pemberian secara mandiri BAP 1,5 ppm terbaik dalam penambahan jumlah daun sebanyak 2,49 buah, BAP 1,5 ppm terbaik dalam memacu pertumbuhan akar sebanyak 1.58 dan BAP 1,5 ppm terbaik dalam meningkatkan tinggi plantlet hingga 0,90 cm. |
| Abtrak (Bhs. Inggris) | The Hybrid orchid Vanda 'Propathom gold' x Vanda 'Robert delight blue' is the result of an easy-to-flower vanda cross with large, beautiful flowers. Through crossbreeding with genetic improvement efforts, it is hoped that it can improve the quality of vanda. To determine the beauty of the interspecific crossed flowers, in vitro culture techniques are needed to increase the quantity. This study was conducted with the aim to determine the effect of IBA and BAP on the growth of Vanda 'Propathom gold' x Vanda 'Robert delight blue' plantlets and to determine the best IBA and BAP interactions for the growth of Vanda 'Propathom gold' x Vanda 'Robert plantlets. delight blue 'blue '. The research was carried out experimentally with the basic design of a completely randomized design (CRD) with a factorial treatment pattern. The first factor is the concentration of IBA (I) with 4 levels, namely I1: 0 ppm, I2: 0.5 ppm, I3: 1 ppm, and I4: 1.5 ppm. The second factor is the concentration of BAP (B) with 4 levels, namely B1: 0 ppm, B2: 0.5 ppm, B3: 1 ppm, and B4: 1.5 ppm. Each treatment combination was repeated 3 times so that there were 48 experimental units. The variables observed were plantlet growth, with parameters measured including the number of leaves, number of roots, and plantlet height. The data obtained were analyzed by Analysis of Variance with a confidence level of 95%, and for treatment that had a real effect, continued with the Duncan Multiple Range Test (DMRT) at the 95% confidence level. The results showed that the best interaction for root growth was IBA 1 ppm and BAP 1.5 ppm as much as 2.11 roots, and for plantlet height increase, namely IBA 1.5 ppm and BAP 1 ppm to 0.98 cm. Independently of 1 ppm IBA was the best in boosting root growth as much as 1.88 roots, and IBA 1.5 ppm was the best in increasing plantlet height up to 0.91 cm. Independently the best BAP 1.5 ppm in increasing the number of leaves as much as 2.49 leaves, the best BAP 1.5 ppm in spurring root growth as much as 1.58 roots, and BAP 1.5 ppm the best in increasing plantlet height to 0.90 cm. |
