Judul ArtikelEFFECTS OF GOTU KOLA (Centella asiatica) EXTRACTS ON CYCLOOXYGENASE-2 (COX-2) LEVELS AND NEUTROPHILS PERCENTAGES IN CADMIUM INDUCED-RAT
Abstrak (Bhs. Indonesia)Kadmium adalah logam berat umumnya melalui proses antropogenik; namun cadmium akan berikatan dengan Metallothionein (Cd-Mt) setelah memasuki tubuh. Sebagai akibatnya, ROS terbentuk, meningkatkan kadar cyclooxygenase-2 (COX-2) dan persentase neutrofil selama proses peradangan. Pemberian ekstrak Pegagan (Centella asiatica) mengandung flavonoid akan menyumbangkan ion-ion H+ untuk menetralkan ROS. Tujuan penelitian adalah mengevaluasi efek dari ekstrak pegagan dan menentukan dosis ekstrak pegagan yang paling efektif pada kadar cyclooxygenase-2 (COX-2) dan persentase neutrofil pada tikus diinduksi kadmium. Metode penelitian menggunakan metode penelitian eksperimental dengan Rancangan Acak Lengkap terdiri (RAL) terdiri dari lima perlakuan dan ulangan. Tikus diindiksi kadmium 25% dari LD50 yaitu 14 mg.kg-1 BB. Tikus secara acak dibagi P0 (Kontrol); P1 (Pegagan 100 mg.kg-1 BB); P2 (Pegagan 200 mg.kg-1 BB); P3 (Pegagan 300 mg.kg-1 BB), dan P4 (Pegagan 400 mg.kg-1 BB). Hasil data penelitian dianalisis menggunakan ANOVA dengan tingkat kepercayaan 5%. Data signifikan dianalisis menggunakan uji post hoc Duncan dengan tingkat kepercayaan 95%. Hasil penelitian menunjukkan pemberian ekstrak Pegagan pada dosis yang berbeda dapat mengurangi kadar cyclooxygenase-2 (COX-2) dan persentase neutrofil pada tikus yang diinduksi kadmium. Semua dosis mempengaruhi penurunan kadar cyclooxygenase-2 (COX-2) dan persentase neutrofil pada tikus yang diinduksi kadmium. Namun, dosis paling efektif yang dapat menurunkan kadar cylooxygenase-2 (COX-2) dan persentase neutrofil mendekati kadar normal adalah dosis 300 mg.kg-1 BB pada tikus putih yang diinduksi kadmium.
Abtrak (Bhs. Inggris)Cadmium is a heavy metal commonly through the anthropogenic processes; however cadmium will bind to Metallothionein (Cd-Mt) after it enters the body. As a consequenced, ROS might form thus, it will increase the cyclooxygenase-2 (COX-2) levels and neutrophils percentages during the inflammation process. The administration of Gotu kola (Centella asiatica) extracts containing flavonoids will donate H+ ions to neutralize ROS. The objectives of this research will evaluate the effects of Gotu kola extracts and determine the most effective dose of Gotu kola extracts on cyclooxygenase-2 (COX-2) levels and neutrophils percentages in cadmium induced-rat. The research method conducted using the experimental research method with a Completely Randomized Design (CRD) consists of five treatments and replications. Rats induce cadmium of 25% of LD50 is 14 mg.kg-1 of BW. Rats randomly was divided into P0 (Controls); P1 (Gotu kola of 100 mg.kg-1 of BW); P2 (Gotu kola of 200 mg.kg-1 of BW); P3 (Gotu kola of 300 mg.kg-1 of BW), and P4 (Gotu kola of 400 mg.kg-1 of BW). The results data analyzed using ANOVA with a significant level of 5%. The significant data has followed by Duncan post hoc test with a confidence level of 95%. The research result shows the administration of Gotu kola extracts at different doses might decreased the cyclooxygenase-2 (COX-2) levels and decreased the neutrophils percentages in cadmium induced-rats. Although all doses have affects reduces cyclooxygenase-2 (COX-2) levels and neutrophils percentages in cadmium induced-rats. Further, it has considered the most effective dose of Gotu kola extracts affected decreases cyclooxygenase-2 (COX-2) levels and neutrophils percentages close to normal levels at 300 mg.kg-1 of BW dose in cadmium induced-rats.
