Penelitian yang bertujuan untuk mengkaji pengaruh penambahan tepung daun sirih pada pakan kambing secara in vitro terhadap konsentrasi N-NH3 dan sintesis protein mikroba rumen. Telah dilaksanakan dari tanggal 11 Desember 2019 sampai 29 Januari 2020 yang bertempat di Laboratorium Ilmu Nutrisi dan Makanan Ternak, Fakultas Peternakan, Universitas Jenderal Soedirman, Purwokerto. Penelitian ini menggunakan metode experimental yang dilakukan secara in vitro dengan rancangan acak lengkap (RAL). Terdapat tiga perlakuan pada penelitian ini, yaitu P0: 40% jerami padi amoniasi + 60% konsentrat + 0% tepung daun sirih, P1: 40% jerami padi amoniasi + 60% konsentrat + 5% tepung daun sirih (berdasarkan BK pakan),P2: 40% jerami padi amoniasi + 60% konsentrat + 10% tepung daun sirih (berdasarkan BK pakan). Setiap perlakuan diulang enam kali sehingga terdapat 18 unit percobaan. Peubah yang diukur yaitu konsentrasi N-NH3 dan sintesis protein mikroba (SPM). Data yang diperoleh kemudian dianalisis menggunakan analisis variansi dan dilanjutkan dengan uji orthogonal polynomial. Rataan konsentrasi N-NH3 yang diperoleh yaitu P0 = 18,33 ± 1,16, P1 = 19,00 ± 0,89, P2 = 20,60 ± 1,25 (Mm); sedangkan rataan untuk sintesis protein mikroba sebesar P0 = 14,91 ± 1,16, P1 = 14,35 ± 2,01, P2 = 11,11 ± 1,41 (mg/20ml). Penambahan tepung daun sirih meningkatkan konsentrasi N-NH3 secara linier dengan persamaan Y= 18,20 + 0,23x dengan koefisien determinasi (r2) sebesar 0,44. Penambahan tepung daun sirih menurunkan sintesis protein mikroba secara linier dengan persamaan Y = 15,46 - 0,38x dengan koefisien determinasi (r2) sebesar 0,50. Kesimpulan dari penelitian ini bahwa penambahan tepung daun sirih pada pakan kambing sampai dengan taraf 10% dapat meningkatkan konsentrasi N-NH3 akan tetapi menurunkan sintesis protein mikroba. |

This study aimed to evaluate the concentration of N-ammonia and protein synthesis of rumen microbes production in goat by supplementing Piper betel leaf flour in the ration. It has been held from December 11, 2019, to January 29, 2020, which is located at the Laboratory of Animal Nutrition and Feed Sciences, Faculty of Animal Science, Jenderal Soedirman University, Purwokerto. This study uses an experimental method with a completely randomized design (CRD). There were three treatments in this study, namely P0: 40% ammoniated rice straw + 60% concentrate + 0% betel leaf flour, P1: 40% ammoniated rice straw + 60% concentrate + 5% betel leaf flour (based on BK feed), P2 : 40% ammoniated rice straw + 60% concentrate + 10% betel leaf flour (based on food BK). Each treatment was repeated six times so that there were 18 experimental units. The variables measured were N-ammonia concentration and microbial protein synthesis. The data obtained were then analyzed using analysis of variance and continued with the polynomial orthogonal test. The average N-ammonia concentration obtained were T0 = 18.33 ± 1.16, T1 = 19.00 ± 0.89, T2 = 20.60 ± 1.25; while the mean for microbial protein synthesis production were T0 = 14.91 ± 1.16, T1 = 14.35 ± 2.01, T2 = 11.11 ± 1.41. The supplementation of Piper betel leaf flour linearly increased the concentration of N-ammonia with the equation Y = 18.20 + 0.23x with a coefficient of determination (r2) of 0.44. The supplementation of betel leaf flour linearly reduced microbial protein synthesis by the equation Y = 15.46 - 0.38x with a coefficient of determination (r2) of 0.50. The conclution that the supplementation of Piper betel leaf flour to goat ration up to a level of 10% can increase the concentration of N-ammonia but reduce microbial protein synthesis. |
