Penelitian ini bertujuan mengetahui karakterisasi isolat bakteri rizosfer padi toleran pestisida sintetik berbahan aktif buprofezin secara morfologi dan biokimia. Penelitian dilaksanakan di Laboratorium Perlindungan Tanaman dan Laboratorium Agroekologi Fakultas Pertanian, Universitas Jenderal Soedirman. Penelitian dilaksanakan sejak bulan November 2018 sampai dengan Februari 2019. Penelitian ini merupakan penelitian eksplorasi menggunakan isolat bakteri SR-1, PA-1, dan GT-2 hasil kulturasi secara murni di mediun nutrient agar. Variabel yang diamati, meliputi karakterisasi morfologi (kelompok positif/negatif dengan pewarnaan gram dan pewarnaan spora) dan karakterisasi biokimia (respon positif/negatif dengan uji katalase, fermentasi karbohidrat, hidrolisis urea, methyl red, voges proskauer, indol, sitrat, motilitas, hidrolisis pati, produksi H2S, oksidasi-fermentasi, dan oksidase). Data hasil penelitian dianalisis secara deskriptif kualitatif yang bertujuan untuk mendeskripsikan karakteristik dari masing-masing isolat bakteri. Hasil penelitian menunjukkan bahwa 1) karakter morfologi dari tiga isolat bakteri tersebut termasuk pada kelompok gram positif, bentuk sel batang, dan memiliki endospora. 2) karakter biokimia yang menunjukkan respon positif pada SRI meliputi uji katalase, fermentasi glukosa, fermentasi manitol, methyl red, motilitas, hidrolisis pati, dan bersifat aerob/anaerob fakultatif. PA1 meliputi uji katalase, methyl red, sitrat, motilitas, produksi H2S, oksidase dan bersifat aerob obligat. GT2 meliputi u ji katalase, fermentasi sukrosa, fermentasi laktosa, fermentasi manitol, methyl red, voges proskuer, indol, sitrat, motilitas, oksidasi-fermentasi, oksidase, dan bersifat aerob/anaerob fakultatif. 3) genus tiga isolat bakteri tersebut berdasarkan Bergeys’s Manual of Determinative Bacteriology 9th adalah Bacillus sp.

This research aims to know morphology and biochemistry character of bacteria isolate on rice plant root that tolerance to synthetic pesticide with buprofezin active compound. This study carried out in Pest and Plant Disease laboratory and Agroecology laboratoty of Agriculture Faculty, Jenderal Soedirman University. The study design is exploration using bacteria isolates SR1, PA1, and GT2 recultured to the agar medium. Variables observed on those 3 (three) bacteria isolates were shape, morphological character (positive/negative group on gram and spore staining) and biochemistry characterization (positive/negative respond on catalase test, carbohydrate fermentation, urea hydrolysis, methyl-red, voges-prostekuer, indole, cytrat, motility, starch hydrolysis, H2S production, fermentation-oxidation, and oxidase). The research data is analysed in a qualitative descriptive way to describe the characteristics of each bacteria isolate. The result of study show that 1) morphology character of the isolates found equation on it’s positive gram, bar shape, and posses endospore. 2) biochemistry character from those isolates that showed positive respond of SR-1 isolate were catalase, glucose and manitol fermentation, methyl red, and motility, starch hydrolysis, and is aerobic/anaerobic facultative. PA-1 were catalase, methyl red, citrate, motility, H2S production, oxidase, and is aerobic obligat. GT-2 were catalase, sucrose, lactose, and manitol fermentation, methyl red, voges proskauer, indole, citrate, motility, oxidase, and is aerobic/anaerobic facultative. 3) genus of three bacteria isolates bassed on Bergeys’s Manual of Determinative Bacteriology 9th is Bacillus sp.
