Abstrak (Bhs. Indonesia)Latar Belakang: Chronic Kidney Disease (CKD) adalah penyakit kerusakan ginjal secara struktural maupun fungsional yang berlangsung selama ≥ 3 bulan. Patomekanisme CKD melibatkan stress oksidatif dan proses inflamasi kronik yang dapat ditandai dengan peningkatan kadar C-Reactive Protein (CRP). Ekstrak etanol seledri (Apium graveolens L.) mengandung zat aktif yang berfungsi sebagai antioksidan dan antiinflamasi sehingga berpotensi mencegah kerusakan pada ginjal.
Tujuan: Penelitian ini bertujuan untuk mengetahui efek pemberian seledri (Apium graveolens L.) dalam mencegah peningkatan kadar CRP tikus putih (Sprague dawley) model CKD.
Desain Penelitian: Metode penelitian adalah eksperimental dengan post test only with control group design. Dua puluh lima ekor tikus dibagi kedalam 5 kelompok yaitu kelompok A sebagai kontrol sehat, kelompok B sebagai kontrol sakit, kelompok C (250 mg/kgBB), kelompok D (500 mg/kgBB), dan kelompok E (1000 mg/kgBB). Pada hari ke 15 setelah pemberian ekstrak, kelompok B, C, D, E dibedah dan dibuat model CKD. Pemberian ekstrak dilanjutkan hingga hari ke 29. Kadar CRP diukur menggunakan Rat HS-CRP Elisa ELISA Kit.
Hasil: Rerata kadar CRP kelompok A=307,14±40,51; B=346,63±34,89; C=212,72±52,81; D=311,23±117,75; E=313,99±92,54. Rerata kadar CRP terendah didapatkan pada kelompok C dan rerata tertinggi pada kelompok B. Hasil uji Kruskal-Wallis rerata kadar CRP menunjukkan nilai p=0,051 (p>0,05).
Kesimpulan: Pemberian ekstrak etanol seledri (Apium graveolens L.) berpotensi untuk mencegah peningkatan kadar CRP tikus model CKD.
Abtrak (Bhs. Inggris)Background: Chronic Kidney Disease (CKD) is define as abnormalities of kidney structure or function that present for ≥ 3 months. The pathomechanism of CKD involving oxidative stress and chronic inflammatory process that can be marked by elevated of C-Reactive Protein (CRP) levels. Ethanol extract of celery (Apium graveolens L.) contains active substances that has antioxidant and anti-inflammatory effect so it can prevent the elevation of CRP levels.
Objective: The aim of this study was to analyze the effect of ethanol extract of celery in preventing the elevation of CRP levels in CKD rats model (Sprague dawley).
Method: This study was an experimental study with post test only control group design. Twenty five male rats were used and divided into 5 groups. Group A as healthy control, group B as unhealthy control, group C (250 mg/kgBW of celery extract), group D (500 mg/kgBW of celery extract), and group E (1000 mg/kgBW of celery extract). On the 15th day after administration of the extract, group B, C, D, and E were dissected and made CKD models. Administration of the extract and aquades was continued until day 29. CRP levels was measured with Rat HS-CRP ELISA Kit.
Result: The mean of CRP on group A=307,14±40,51; B=346,63±34,89; C=212,72±52,81; D=311,23±117,75; E=313,99±92,54. The lowest mean of CRP level is found on Group C and the highest CRP level is found on Group B. Kruskal-Wallis test showed p value= 0,051 (p>0,05).
Conclusion: Ethanol extract of celery has a potential effect to prevent elevation of CRP levels.
