| Abstrak (Bhs. Indonesia) | Gingivitis merupakan peradangan atau inflamasi pada gingiva yang disebabkan oleh penumpukan plak yang mengandung bakteri seperti Porphyromonas gingivalis. Selama inflamasi terjadi aktivitas berlebih sel neutrofil yang menyebabkan keadaan stress oksidatif yang memicu kerusakan jaringan. Intracellular adhesion molecule (ICAM-1) merupakan molekul transmembran protein yang terekspresi ke permukaan sel endotel, fibroblast dan epitel apabila terjadi keadaan inflamasi yang disebabkan oleh infeksi bakteri. Daun pepaya memiliki senyawa aktif seperti flavonoid, papain dan tannin yang dapat berperan sebagai antioksidan eksogen. Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak daun pepaya terhadap kadar ICAM-1 pada tikus wistar model gingivitis. Jenis penelitian ini adalah eksperimental laboratoris dengan posttest-only control group design menggunakan 30 ekor tikus Rattus norvegicus jantan yang dibagi mejadi 5 kelompok, yaitu kelompok kontrol sehat (KS), kontrol negatif (KN), perlakuan pemberian ekstrak konsentrasi 50% (KP1), 75% (KP2), 100% (KP3). Kelompok perlakuan dan kontrol negatif diinduksi P. gingivalis 0,02 ml pada gingiva gigi insisivus rahang bawah selama 5 hari berturutan. Ekstrak diberikan per oral 0,05 ml pagi hari selama 6 hari (kelompok perlakuan). Setiap kelompok diambil supernatan jaringan gingiva insisivus rahang bawah dan dilakukan pemeriksaan kadar ICAM-1 menggunakan ELISA. Data dianalisis menggunakan One-way ANOVA dan Post hoc LSD. Hasil penelitian menunjukkan terdapat perbedaan kadar ICAM-1 sangat bermakna antara kelompok kontrol negatif dengan seluruh kelompok perlakuan (p<0,01), sementara tidak terdapat perbedaan kadar ICAM-1 yang bermakna kelompok kontrol sehat dengan kelompok ekstrak konsentrasi 75% dan 100%. Pemberian ekstrak etanol daun pepaya memiliki pengaruh terhadap penurunan kadar ICAM-1 pada tikus Wistar model gingivitis dengan konsentrasi efektif pemberian ekstrak yaitu 75%. |
| Abtrak (Bhs. Inggris) | Gingivitis is an inflammation of the gingiva caused by plaque accumulation containing bacteria such as Porphyromonas gingivalis. During the inflammation process, excessive activity of neutrophil cells causes oxidative stress that promotes tissue damage. Intracellular adhesion molecule (ICAM-1), a protein transmembrane molecule, expressed to the surface of endothelial, fibroblast, and epithelial cells during inflammatory condition caused by bacterial infection. Papaya leaf has active compound such as flavonoid, papain, and tannin which act as exogenous antioxidant. The aim of this study was to determine the effect of papaya leaf extract to ICAM-1 level in gingivitis wistar rat model. This laboratory experiment with post test-only control group design used 30 male Rattus norvegicus. The subject of this research were divided into 5 groups, i.e. healthy control (KS), negative control (KN), treatment group 50% (KP1), 75% (KP2), and 100% (KP3). The treatment group and negative control were induced with 0,02 ml P. gingivalis in the mandible incisor consecutve for 5 days. Extract was given orally for 0,05 ml in the morning for 6 days (treatment group). Gingival tissue from each groups were collected from mandible incisor and ICAM-1 level was examined using ELISA. Data were analyzed using one-way ANOVA and post-hoc LSD. The results of this study showed that there was a significant difference of ICAM-1 level between negative control group and all treatment groups (p<0,01). Meanwhile, there was no significant difference of ICAM-1 level between healthy control group and 75% and 100% treatment group. The papaya leaf ethanol extract contributes to the decrease of ICAM-1 level in gingivitis wistar rat model with effective concentration is 75%. |
