| Abstrak (Bhs. Indonesia) | Penelitian ini bertujuan mengetahui tanggap embriogenesis dari beberapa aksesi cabai koleksi PT. East West Seed Indonesia melalui kultur antera menggunakan media dua-lapis. Penelitian dilaksanakan di Laboratorium Biologi Sel PT. East West Seed Indonesia, pada bulan Maret 2018 sampai dengan bulan Agustus 2018. Rancangan percobaan yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan 6 taraf yaitu, aksesi cabai (Ewindo 1-6) dengan 12 ulangan. Setiap perlakuan menggunakan 4 cawan petri dan setiap cawan petri terdiri dari 5 bunga. Data hasil penelitian dianalisis menggunakan Least Significant Difference (LSD) pada taraf nyata 5%. Bunga yang dipanen adalah bunga yang mengandung 50% mikrosporanya berada pada stadia uninukleat akhir, kemudian bunga di sterilisasi. Kuncup bunga diberi praperlakuan 4oC selama satu hari sebelum diisolasi. Antera yang sudah dipisahkan dari kelopak, mahkota dan putiknya dikultur pada media dua-lapis kemudian diinkubasi pada suhu 9oC selama 7 hari. Inkubasi kemudian dilanjutkan pada suhu 28oC sejak 3 minggu setelah kultur (msk) sampai dengan 10 msk, pada minggu ke 3 media ditambahkan zpt berupa IAA 0,05 ppm dan zeatin 0,03 ppm. Hasil penelitian menunjukkan bahwa induksi embriogenesis terjadi pada seluruh aksesi dengan variasi yang beragam. Aksesi Ewindo 5 (A5) adalah aksesi yang paling responsif dengan rata-rata embrio per kuncup mencapai 3,97 kemudian membutuhkan waktu rata-rata induksi tersingkat yaitu 51,33 hsk dengan persentase embrio normal sebesar 43%. Aksesi lainnya menunjukkan respon yang beragam terhadap setiap variabel yang diamati. Aksesi A2 hanya mampu menginduksi 0,03 embrio per kuncup bunga, namun embrio yang dihasilkan 60%nya adalah embrio normal, sehingga aksesi ini adalah aksesi dengan normalitas tertinggi. |

| Abtrak (Bhs. Inggris) | This study aimed to determine embryogenesis response from several accessions of hot pepper from PT. East West Seed Indonesia collections by anther culture with double layered medium. The research was carried out at Biology Cell Laboratory, PT. East West Seed Indonesia, started from March 2018 to August 2018. The experimental design used was a Completely Randomized Design (CRD) with 6 levels, namely, hot pepper accession (Ewindo 1-6) with 12 replications. Four petri dishes were used in each treatment and each petri dish consists of 5 flowers. Data were analyzed using Least Significant Difference (LSD) at a significant level of 5%. The flower harvested is a flower containing 50% of the microspores in the late uninucleate stage, then the flowers were sterilized. Flower buds were pre-treated with 4oC for one day before being isolated. Anther which has been separated from the petals, crown and pistil was stored in a double layered medium and then incubated at 9oC for 7 days. The results of the culture were then incubated at 28oC from 1 st to 10 week after culture, in the third week after cultured the media was added by growth regulator in the form of IAA 0.05 ppm and zeatin 0.03 ppm. The results showed that embryogenesis induction occurred in all accessions with various variations. Ewindo 5 (A5) accession is the most responsive accession with the average embryo per bud reaching 3.97 then requiring the shortest average induction time of 51.33 days with a normal percentage of 43% embryos. Other accessions show diverse responses to each observed variable. Accession A2 is only able to induce 0.03 embryos per flower bud, but the embryo produced by 60% are normal embryos, so this accession is the accession with the highest normality. |
