Abstrak (Bhs. Indonesia)Penelitian ini bertujuan untuk mendapat isolat bakteri endofit batang dan daun cabai merah, mengetahui karakteristiknya melalui uji fisiologis dan biokimia, pengamatan secara makroskopis dan mikroskopis, mengetahui daya hambat bakteri endofit terhadap pertumbuhan jamur patogen bercak daun secara in vitro, dan mengetahui daya hambat bakteri endofit terhadap perkembangan penyakit bercak daun cabai secara in planta. Penelitian ini dilaksanakan di Laboratorium Perlindungan Tanaman dan Screen House Fakultas Pertanian, Universitas Jenderal Soedirman, Purwokerto mulai bulan Oktober sampai dengan Juli 2018. Penelitian ini diawali dengan isolasi dan identifikasi bakteri endofit asal batang dan daun tanaman cabai merah. Uji in vitro menggunakan Rancangan Acak Lengkap (RAL) dengan 8 perlakuan dan 3 ulangan terdiri atas perlakuan CB0 (kontrol), CB1 (isolat 1), CB2 (isolat 2), CB3 (isolat 3), CB6 (isolat 6), CB7 (isolat 7), CB8 (isolat 8), dan CB9 (isolat 9). Pengujian in planta menggunakan Rancangan Acak Kelompok Lengkap(RAKL) dengan 6 perlakuan dan 4 ulangan terdiri atas perlakuan CB0 (kontrol), CB1 (patogen + isolat 1), CB2 (patogen + isolat 2), CB3 (patogen + isolat 3), CB6 (patogen + isolat 6) dan CB7 (patogen + isolat 7). Variabel yang diamati macam bakteri endofit, masa inkubasi, intensitas penyakit, laju infeksi, tinggi tanaman dan jumlah daun. Hasil penelitian ini menunjukan bahwa bakteri endofit batang dan daun cabai yang didapatkan terdiri atas 7 isolat bakteri yaitu 3 isolat bakteri endofit berasal dari batang dan 4 isolat berasal dari daun tanaman cabai merah. Bakteri isolat 1 dan isolat 3 asal batang, serta isolat 8 dan isolat 9 asal daun belum teridentifikasi. Bakteri isolat 2 asal batang teridentifikasi memiliki karakteristik genus Bacillus sp. sedangkan isolat 6 dan isolat 7 memiliki karateristik bakteri genus Arthrobacter sp. Berdasarkan uji in vitro bakteri endofit batang dan daun dapat menghambat pertumbuhan patogen sampai dengan 14,47%. Hasil uji in planta menunjukan bakteri endofit isolat 2 dapat menunda masa inkubasi dan menekan penyakit bercak daun sebesar 26,59%, namun belum efektif untuk menghambat perkembangan penyakit tersebut. Perlakuan 5 isolat bakteri endofit asal batang dan daun secara in planta belum mampu meningkatkan tinggi tanaman dan jumlah daun.

Inggris)This research aims to determine the endophytic bacteria isolates of chilli stem and leaf, to know its characteristics through physiological and biochemical test, macroscopic and microscopic test, to know endophytic bacteria inhibition to growth of the patogenic fungi leaf spot using in vitro testing and to know endophytic bacteria inhibition to development of leaf spot disease using in planta testing. This research was conducted at the Plant Protection Laboratory and Screen House, Faculty of Agriculture, Jenderal Soedirman University, Purwokerto, start in October until July 2018. The research started with isolation and identification of endophytic bacteria from stem and leaves of red chili. in vitro test using Completely Randomized Design with 8 treatments and 3 replications consist of control treatment CB0 (control), CB1 (isolate 1),CB2 (isolate 2), CB3 (isolate 3), CB6 (isolate 6), CB7 (isolate 7), CB8 (isolate 8) and CB9 (isolate 9). in planta test using Rendomized Complete Block Design with 6 treatments and 4 replications consist of CB0 (control), CB1 (pathogen + isolate 1), CB2 (pathogen + isolate 2), CB3 (pathogen + isolate 3), CB6 ( pathogen + isolate 6) and CB7 (pathogen + isolate 7). Observed variables in this research is endophytic bacteria, incubation period, disease intensity, infection rate, plant height and number of leaves. Result of the research showed that endophytic bacteria of chilli stem and leaves were obtained 7 endophytic bacteria. 3 isolates from stem and 4 isolates from leaves. Bacteria isolate 1 and isolate 3 from stem, isolate 8 and isolate 9 from leaves has not been identified. Isolate 2 from stem were identified as have characteristic of the genus Bacillus sp. whereas isolate 6 and 7 have characteristic of the genus Arthrobacter sp. Based on in vitro test, endophytic bacteria of stems and leaves were able to inhibit pathogen growth to 14,47%. ‘In planta’ test results showed that endophytic bacteria isolate 2 can delayed incubation period and suppress leaf spot disease by 26,59%, but it has not been effective to inhibit the development of disease. Treatment with in planta test of 5 isolates endophytic bacteria from stem and leaves has not been able to increase plant height and leaf number.
