Indonesia) | Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi beberapa bakteri endofit akar tomat asal Kecamatan Baturraden, mengisolasi dan mengidentifikasi bakteri layu asal tanaman tomat, mengetahui pengaruh bakteri endofit akar tomat dalam menekan pertumbuhan bakteri layu secara in-vitro, dan mengetahui mekanisme penghambatan bakteri endofit terhadap bakteri layu. Penelitian dilaksanakan di lahan pertanaman tomat Kecamatan Baturraden dan Laboratorium Perlindungan Tanaman, Fakultas Pertanian, Universitas Jenderal Soedirman pada bulan November 2017 sampai April 2018. Pengujian in-vitro menggunakan Rancangan Acak Lengkap dengan 6 perlakuan dan 4 ulangan, meliputi kontrol (tanpa bakteri endofit) dan 5 isolat bakteri endofit. Variabel yang diamati meliputi uji morfologi, gram, katalase, hidrolisis pati, pertumbuhan pada beberapa pH, suhu dan konsentrasi NaCl, zona hambatan, mekanisme antagonis, dan kurva pertumbuhan bakteri endofit. Hasil penelitian menunjukkan bahwa bakteri endofit telah diisolasi dari akar tanaman tomat asal Kecamatan Baturraden dan didapatkan 5 isolat yang terdiri IN1, IN2, IN3, IN4, dan IN5. Isolat IN1, IN4, dan IN5 termasuk bakteri kelompok Bacillus sp., isolat IN3 termasuk bakteri Pseudomonas kelompok fluorescens, sedangkan isolat IN2 tidak termasuk bakteri Bacillus sp. atau Pseudomonas kelompok fluorescens. Penyebab penyakit layu bakteri terindentifikasi sebagai R. solanacearum. Bakteri endofit isolat IN4 asal Karang Tengah memiliki tingkat penghambatan paling besar terhadap R. solanacearum dengan zona hambatan sebesar 6,55 mm (metode inokulasi titik), 4,00 mm (metode paper disk), dan 6,30 mm (metode sumuran). Mekanisme penghambatan bakteri endofit terhadap R. solanacearum adalah bakteriostatik. |

Inggris) | This research was aimed to isolation and identification some bacterial endophytes from tomato roots in Baturraden sub-distric, to isolation and identification bacterial wilt from tomato plant, to determine the influence of bacterial endophytes from tomato roots in suppressing the growth of bacterial wilt for in-vitro test, and to determine the inhibiton mechanism of endophytes bacterial against bacterial wilt.The research was conducted in tomato cultivation area of Baturraden sub-distric and the Laboratory of Plant Protection of Agriculture, Jenderal Soedirman University from November 2017 to April 2018. Randomized completely design was used for in vitro test with 6 treatments and 4 replication consisted of control (without bacterial endophytes) and 5 isolates of bacterial endophytes. Variables observed were morphological test, gram, catalase, starch hydrolysis, growth at several pH, temperature, and NaCl concentration, inhibition zone, antagonistic mechanism, and endophytic bacterial growth curve. Result of the research showed that bacterial endophytes have been isolated from root of tomato plants from Baturraden sub-district and 5 isolates were found IN1, IN2, IN3, IN4, and IN5. The isolates of IN1, IN4, and IN5 was Bacillus sp., IN3 was Pseudomonas fluorescens group, and IN2 was not identified as Bacillus sp. or Pseudomonas fluorescens group. The bacterial wilt disease was identified as R. solanacearum. Bacterial endophytes isolates IN4 from Karang Tengah showed the highest level of R. solanacearum inhibitation with inhibition zone of 6,55 mm by inoculation method, 4,00 mm by paper disk method, and 6,30 by well diffusion method. The inhibition mechanism of bacterial endophytes against R. solanacearum was bacteriostatic. |
