| Abstrak (Bhs. Indonesia) | Purwoceng merupakan tanaman obat asli Indonesia yang berkhasiat meningkatkan vitalitas pria. Akar purwoceng mengandung flavonoid, alkaloid, saponin, dan tanin yang bersifat lipofilik sehingga berpotensi toksik terhadap ginjal. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak etanol akar Purwoceng (Pimpinella pruatjan Molk.) secara kronik terhadap struktur histologi tubulus proksimal tikus (Rattus norvegicus) jantan. Metode penelitian ini adalah analitik eksperimental dengan post-test only with control group design. Intervensi dilakukan selama 90 hari pada 24 ekor tikus yang terbagi ke dalam 4 kelompok, yaitu kelompok kontrol (A), kelompok perlakuan dengan dosis 21 mg/kgBB (B), 42 mg/kgBB (C), dan 84 mg/kgBB (D). Kerusakan struktur histologi ginjal dihitung berdasarkan jumlah atrofi tubulus, nekrosis tubulus, infiltrasi limfosit, dan fibrosis intersisial. Rerata total skor kerusakan ginjal kelompok A=1,80 ± 0,67, B=2,23 ± 0,39, C= 5,23 ± 0,85 dan D= 6,97± 0,79. Analisis data menunjukkan terdapat perbedaan kerusakan histologi ginjal yang bermakna (One Way ANOVA ). Hasil uji Post Hoc LSD menunjukkan perbedaan bermakna antara kelompok A dengan kelompok C dan D, kelompok C dengan D (p<0,05). Perbedaan kerusakan tidak bermakna ditemukan pada kelompok A dengan B (p=0,296). Pemberian ekstrak etanol akar purwoceng secara kronik berpengaruh terhadap struktur histologi tubulus proksimal, dengan dosis toksik minimal 42 mg/kgBB dan dosis aman yang dapat digunakan 21 mg/kgBB. |

| Abtrak (Bhs. Inggris) | Purwoceng is one of Indonesia indigenous plants used for increasing male vitality. Root of purwoceng comprises of flavonoid, alkaloid, saponin, and tannin which are lipophilic and potentially toxic against the kidney. The purpose of this research is to find out the chronic effect of ethanol-purwoceng root extract administration to the histologic structure of renal proximal tubules in male rats (Rattus norvegicus). This research was an analytical experiment with post-test only with control group design method. Intervention was given for 90 days to 24 male rats that were distributed in 4 groups. The groups consist of a control group, groups with the administration of extract for 21 mg/kgBW (B), 42 mg/kgBW (C), and 84 mg/ kgBW (D). The damage of histologic renal structure was calculated based on the amount of tubules atrophy, tubules necrosis, lymphocytes infiltration, and interstitial fibrosis. Means of renal damage total score of group A=1.80 ± 0.67, B=2.23 ± 0.39, C= 5.23 ± 0.85 and D= 6.97± 0.79. The data analysis indicates that there is a significant difference of histological kidneys damaged (One Way ANOVA). The result of Post Hoc LSD method of analysis showed a significant difference between group A against group C and D, group C and D (p<0.05). There was no significant difference of renal damage found between group A and group B (p=0.296). The chronics administration of purwoceng roots ethanol extract influences to the histological structure of renal proximal tubules with the minimal toxic dose is 42 mg/kgBW and the safe dose that can be used is 21 mg/kgBW. |
