| Abstrak (Bhs. Indonesia) | Latar Belakang: Daun Sendok (Plantago major l.) merupakan tanaman obat (herbal) yang berguna sebagai hepatoprotektor, antioksidan, antiapoptosis dan antiinflamasi. Senyawa daun sendok berupa tannin, aucubin, flavonoid, terpenoid, dan alkaloid berpotensi menimbulkan efek toksik pada tubuh. Uji toksisitas akut oral merupakan salah satu cara untuk mengetahui bagaimana pengaruh suatu zat toksik terhadap tubuh dalam jangka waktu satu hari. Indikator yang dapat digunakan dalam menilai tingkat toksisitas suatu zat adalah gambaran histopatologi hepar. Tujuan: Tujuan dari penelitian ini adalah untuk mengetahui bagaimana pengaruh pemberian ekstrak etanol daun sendok (Plantago major l.) terhadap gambaran histopatologi tikus putih (Rattus norvegicus) jantan galur Wistar pada uji toksisitas akut. Metode: Penelitian ini bersifat eksperimental dengan pendekatan post-test only terhadap tikus putih jantan galur Wistar. Intervensi dilakukan terhadap 24 ekor tikus yang dibagi kedalam empat kelompok secara acak, yaitu kelompok kontrol (A), kelompok perlakuan 1 dengan dosis 1000mg/kgBB, (B), kelompok perlakuan 2 dengan dosis 2000mg/kgBB (C), dan kelompok perlakuan 3 dengan dosis 4000mg/kgBB (D). Tingkat kerusakan histopatologi hepar dinilai dengan menggunakan skoring Manja Roegnik. Hasil: Rerata skor Manja Roegnik kelompok A=1.6218±0.57166, kelompok B=2.46±0.62680, kelompok C=2.6333±0.26493, dan kelompok D=2.0400±0.82185. Uji Oneway-Anova menunjukkan terdapat perbedaan kerusakan gambaran histopatologi yang bermakna (p=0.049). Uji Post-hoc menunjukkan perbedaan bermakna (p<0.05) pada perbandingan kelompok A dengan kelompok C (p=±1.01). Perbedaan tidak bermakna (p>0.05) ditemukan pada perbandingan kelompok A dengan kelompok B (p=±0.83) dan D (p=±0.41), kelompok B dengan kelompok C (p=±0.17) dan D (p=±0.42), serta kelompok C dengan kelompok D (p=±0.59). Kesimpulan: Pemberian ekstrak etanol Daun Sendok secara akut memberikan kerusakan maksimal hepar tikus putih jantan pada dosis 2000mg/kgBB. |
| Abtrak (Bhs. Inggris) | Background: Broadleaf Plaintain (Plantago major 1.) is a herbs of medicinal plant that is used for hepatoprotector, anti-oxidants, anti-apoptosis and anti-inflammation. Broadleaf Plaintain contains tannin, aucubin, flavonoid, terpenoid, and alkaloid with potentially inflicting toxic effect for body. Examine the oral acute toxicity is one of the way to know how the effect of toxic substance on the body within a day. Indicates that it is used to measure the level of the toxicity of substance is an illustrate from histopathology of the liver. Objective: The purpose of this research is to find out how the effect of ethanol extract of Broadleaf Plaintain (Plantago major 1.) with liver histopathological appearance: acute toxicity test. Metode: This research is an experimental approach with post-test only with the male white rat (Rattus Norvegicus). The intervention had been done to 24 rats which were divided into four groups at randomly, Control group (A), treatment group 1 with dosage 1000mg/kgBW (B), treatment group 2 with dosage 2000mg/kgBW (C), and treatment group3 with dosage of 4000mg/kgBW. Histopathologic liver damage rates were assessed using the Manja Roegnik Score. Result: The average score of Manja Roegnik group A = 1.6218±0.571166, group B = 2.46±0.62680, group C = 2.6333±0.26493, and group D = 2.0400±0.82185. The measure of Anova-Oneway showed a significant difference in histopathologic illustrate defects (p=0.049). Post-hoc test showed significant difference in differences (p<0.05) in comparison between group A and group C (p=±1.01). The differences were not significant (p>0.05) found in comparison between group A with group B (p=±0.83) and D (p=±0.41), group B with group C (p=±0.17) and D (p=±0.42), and group C with group D (p=±0.59). Conclution: Giving an extract ethanol of Broadleaf Plaintain continuously damaging the rat male hepar at dosage 2000mg/kbBW Maximally. |
