Email: athoulloh1616@gmail.com ABSTRAK Periodontitis merupakan inflamasi kronik pada jaringan periodontal yang ditandai dengan adanya kehilangan perlekatan (attachment loss) dan kehilangan tulang alveolar (bone loss) sehingga dapat menyebabkan kehilangan gigi pada tahap lanjut. Salah satu kelainan sistemik yang dapat memperparah periodontitis adalah penyakit osteoporosis. Penatalaksanaan dari periodontitis yang disertai osteoporosis biasanya dilakukan terapi konvensional. Terapi konvensional memiliki banyak kekurangan sehingga dibutuhkan terapi alternatif yaitu dengan pemberian simvastatin. Simvastatin merupakan obat yang memiliki efek pleitropik. Tujuan penelitian ini adalah mengetahui pengaruh pemberian simvastatin secara topikal terhadap kadar osteokalsin gingival crevicular fluid tikus model periodontitis dengan osteoporosis. Jenis penelitian ini adalah eksperimental laboratoris dengan rancangan post-test only control group design. Teknik pengambilan sampel adalah metode simple random sampling. Penelitian ini menggunakan 35 sampel yang dibagi menjadi 5 kelompok yaitu kelompok kontrol sehat, kelompok kontrol negatif yang diberi Na-CMC 1% dan 3 kelompok perlakuan yang diberi simvastatin konsentrasi 1,25%, 2,5%, dan 5%. Subjek penelitian adalah tikus Sprague dawley berkelamin betina berumur 2-3 bulan. Tikus diinduksi peridontitis dengan injeksi bakteri A. actinomicetemcomitans, osteoporosis dengan ovariektomi, diberi perlakuan selama 14 hari, dan dilakukan pengukuran kadar osteokalsin GCF menggunakan uji ELISA. Data dianalisis menggunakan uji Saphiro Wilk, uji Levene, One-Way ANOVA dan Post-Hoc LSD. Hasil penelitian menunjukkan kadar osteokalsin pada kelompok perlakuan simvastatin 2,5% dan 5% lebih tinggi dibandingkan kelompok control negatif(p<0,05). Simpulan dari penelitian ini adalah pemberian simvastatin secara topikal dengan konsentrasi 5% dapat meningkatkan kadar osteokalsin GCF tikus model periodontitis dengan osteoporosis. Email: athoulloh1616@gmail.com ABSTRACT Periodontitis is a chronic inflammation of periodontal tissue characterized by attachment loss and alveolar bone loss that can lead to tooth loss in later stages. One of the systemic disorders that can aggravate periodontitis is osteoporosis. The association of osteoporosis with periodontitis is closely related to estrogen hormone deficiency that causes bone damage. Management of periodontitis with osteoporosis is usually done by conventional therapy. Conventional therapy has many disadvantages so that alternative therapy is required by giving simvastatin. Simvastatin is a drug that has pleitropic effects. The purpose of this study was to investigate the effect of topical simvastatin on the osteocalcin level of gingival crevicular fluid periodontitis with osteoporosis rat model. This type of research was experimental laboratory with posttest-only control group design. sampling technique was simple random sampling method. Thirty five samples were divided into 5 groups: healthy control group, negative control group given Na-CMC 1%, and 3 treatment groups given simvastatin concentration 1.25%, 2.5%, and 5%. The subjects of the study were female, 2-3 month-old Sprague dawley rat. Rats were peridontitis induced with injection of A. Actinomicetemcomitans for 7 days and osteoporosis induced with ovariectomy for 14 days. Gingival crevicular fluid osteocalcin level were measured by ELISA. Data were analyzed using Saphiro Wilk test, Levene test, One-Way ANOVA, and Post-Hoc LSD higher The results showed that the osteocalcin level was higher in the 2.5% and 5% simvastatin treatment groups than in the negative control group (p <0.05). Conclusions from this study was topical simvastatin concentration 5% increases the osteocalcin content of gingival crevicular fluid in periodontitis with osteoporosis rat model.
