Judul ArtikelPENGARUH LAMA POST THAWING DAN BANGSA SAPI YANG BERBEDA TERHADAP ABNORMALITAS DAN INTEGRITAS MEMBRAN SEL SPERMATOZOA STRAW SEMEN BEKU
Abstrak (Bhs. Indonesia)Penelitian ini bertujuan untuk mengetahui pengaruh lama post thawing dan bangsa sapi yang berbeda terhadap abnormalitas dan integritas membran sel spermatozoa. Materi yang digunakan dalam penelitian adalah 36 straw yang terdiri dari 12 straw Simmental, 12 straw Brahman, dan 12 straw Limousine. Larutan yang digunakan adalah larutan eosin-nigrosin, nitrogen cair dan larutan HOS (Hypo Osmotic Swollen) test. Penelitian dilakukan dengan metode eksperimental yang dirancang menggunakan rancangan acak lengkap (RAL) dengan pola faktorial. Perlakuan yang diuji yaitu A1= lama post thawing 20 menit, A2= lama post thawing 40 menit, A3= lama post thawing 60 menit, B1= straw sapi Simmental, B2= straw sapi Brahman, dan B3= straw sapi Limousine. Peubah yang diukur adalah abnormalitas dan integritas membran sel spermatozoa. Nilai persentase abnormalitas dihitung dengan jumlah spermatozoa abnormal dibagi jumlah spermatozoa yang diamati dikali 100%, sedangkan nilai persentase integritas membran sel spermatozoa dihitung dengan cara jumlah spermatozoa dengan ekor melingkar dibagi jumlah sel spermatozoa yang diamati kemudian dikali 100%. Hasil penelitian abnormalitas pada lama post thawing 20, 40, dan 60 menit yaitu 17,58 ± 2,1; 37,52 ± 3,0; dan 64,84 ± 4,7 dan integritas membran sel spermatozoa pada lama post thawing 20, 40, dan 60 menit yaitu 57,88 ± 4,3; 33,58 ± 4,3; dan 20,46 ± 3,0. Hasil analisis variansi menunjukkan bahwa tidak ada interaksi antara lama post thawing dengan bangsa sapi yang berbeda terhadap abnormalitas dan integritas membran sel spermatozoa, namun lama post thawing yang berbeda berpengaruh sangat nyata (P<0,01) terhadap abnormalitas dan integritas membran sel spermatozoa, sedangkan bangsa sapi yang berbeda tidak berpengaruh nyata (P>0,05) terhadap abnormalitas dan integritas membran sel spermatozoa. Berdasarkan hasil penelitian dapat disimpulkan bahwa semakin lama waktu post thawing akan meningkatkan persentase abnormalitas dan menurunkan persentase integritas membran sel spermatozoa.
Abtrak (Bhs. Inggris)The aim of this research was to find out the effect of different post thawing time and cattle breeds on spermatozoa morphological abnormality and membrane integrity. This research used 36 straws, which was 12 Simmental bull straws, 12 Brahman bull straws, 12 Limousine bull straws, eosin-nigrosin solution, liquid nitrogen and HOS test solution. This research used experimental method and that was designed using a completely randomized design (CRD) with factorial pattern. The tested treatments were A1= 20 minutes of post thawing time, A2= 40 minutes of post thawing time, A3= 60 minutes of post thawing time, B1= Simmental’s bull straw, B2= Brahman’s bull straw, and B3= Limousine’s bull straw. The measured variables were the spermatozoa morphological abnormality and membrane integrity. The spermatozoa morphological abnormality percentage were calculated with number of observed abnormal spermatozoa divided with total of observed spermatozoa and then multiplied by 100%, while membrane integrity percentage were calculated with number of observed spiral tailed spermatozoa divided with total of observed spermatozoa and then multiplied by 100%. The average spermatozoa abnormality percentage of 20, 40, and 60 minutes post thawing time is 17.58 ± 2.1, 37.52 ± 3.0, and 64.84 ± 4.7, respectively. The average membrane integrity for 20, 40, and 60 minutes post thawing time is 57.88 ± 4.3, 33.58 ± 4.3, and 20.46 ± 3.0, respectively. The research showed that there was no interaction effect (P>0.05) between different post thawing time and cattle breeds on spermatozoa morphological abnormality and membrane integrity. Different post thawing time had significant effect (P<0.01) on spermatozoa morphological abnormality and membrane integrity, however different cattle breeds did not affect (P>0.05) spermatozoa morphological abnormality and membrane integrity. This research was concluded that extending post thawing time could increase spermatozoa morphological abnormality percentage and decrease membrane integrity percentage of frozen semen.
