| Abstrak (Bhs. Indonesia) | Isolasi dan karakterisasi aktinomisetes penghasil enzim selulase telah dilakukan menggunakan sampel sedimen dari 10 titik sampling di area serasah mangrove Segara Anakan Cilacap. Metode penelitian meliputi isolasi marine aktinomisetes, uji potensi selulolitik (CMC agar - Congo Red), dan identifikasi spesies (uji sifat Gram, uji katalase, pengamatan morfologi koloni, gen 16S rRNA). Hasil isolasi menghasilkan 16 isolat marine aktinomisetes berdasarkan pengamatan morfologi. Delapan isolat aktinomisetes positif menghasilkan enzim selulase dengan Indeks Aktifitas Selulase tertinggi adalah 0,350 (isolat P-6G) dan terendah adalah 0,183 (isolat P-7F). Identifikasi spesies menunjukkan bahwa P-6G bersifat gram (+), berbentuk sel batang, positif pada uji katalase, hifa berbentuk spiral dan berseptat yang mengindikasikan genus Streptomyces. |

Inggris) | Isolation and characterization of cellulase producing actinomycetes have been performed using sediment samples from 10 sampling points in mangrove area of Segara Anakan Cilacap. The methods applied were isolation of marine actinomycetes, cellulase activity test (CMC agar - Congo Red), and species identification (Gram properties test, catalase test, colony morphological observation, 16S rRNA gene). The isolation result yielded 16 isolate marine actinomycetes based on morphological observation. Eight positive actinomycetes isolates produced the cellulase enzyme with the highest Cellulase Activity Index was 0.350 (P-6G isolate) and the lowest was 0.183 (P-7F isolate). The species identification indicated that P-6G is Gram (+), rod-shaped, positive in catalase, spiral with segmented hyphae that indicate the genus of Streptomyces. |
