Abstrak (Bhs. Indonesia)Pestisida telah digunakan secara luas dalam bidang pertanian melalui pembasmian hama. Sifat penting yang dimiliki suatu bahan aktif pestisida adalah daya racun atau toksisitas. Endosulfan (organoklorin pestisida) adalah polutan yang sangat beracun dan persisten yang dapat mencemari perairan sungai. Dampak endosulfan bagi biota perairan menghambat reproduksi terutama ekspresi gen GtH II-α. Biota uji yang digunakan yaitu ikan nilem. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh endosulfan (organoklorin pestisida) terhadap ekspresi gen GtH II-α serta konsentrasi endosulfan (organoklorin pestisida) yang dapat mengganggu ekspresi gen GtH II-α pada ikan nilem (Osteochilus hasselti C.V) betina. Metode yang digunakan adalah metode eksperimental, dengan desain penelitian Rancangan Acak Lengkap (RAL). Penelitian dilakukan dengan 4 tahap, pengambilan hipotalamus, isolasi RNA, DNAse Treatment, Pengukuran konsentrasi RNA dan Real-Time PCR. Sampel diberi 4 perlakuan endosulfan berbeda selama 30 hari dengan pengambilan sampel setiap 2 minggu sekali. Data kuantitatif berupa ekspresi gen penghasil GtH II-α dianalisis dengan one ways Anova dan Uji BNT. Hasil penelitian menunjukkan bahwa pengaruh perlakuan endosulfan selama 30 hari terhadap nilai konsentrasi ekspresi gen GtH II-α memiliki kisaran rata-rata 0,002-2,751. Pengaruh perlakuan endosulfan berupa penurunan nilai konsentrasi ekspresi gen GtH II-α dan nilai IKG, seiring dengan meningkatnya konsentrasi endosulfan serta lamanya waktu pemaparan.

Abtrak (Bhs. Inggris)Pesticides have been widely used in agriculture through the eradication of pests. The essential properties of a ingredient are toxicity. Endosulfan is highly toxic pollutants and persistent that could contaminate the river waters. The Impact of endosulfan for aquatic organism primarily to lower reproductive such as GtH II-α gene expression. Osteochilus hasselti is object for gene expression test. The purpose of this study was to determine the effect of endosulfan on gene expression GtH II-α and concentrations of endosulfan that can interfere with GtH II-α gene expression in Osteochilus hasselti females. The method used was experimental method, the research design Completely Randomized Design. The study was conducted with 4 stages, making the hypothalamus, RNA isolation, DNAse treatment, RNA concentration measurements and Real-Time PCR. Samples were given four different treatment of endosulfan for 30 days with sampling every 2 weeks. The quantitative data in the form of GtH II-α-producing gene expression was analyzed by one-ways ANOVA and BNT test. The results showed that the treatment effect of endosulfan for 30 days against the value of the concentration of gene expression GtH II-α have an average range of 0,002 to 2,751. Endosulfan treatment effect of pesticide in the form of a decrease in the concentration of GtH II-α gene expression and value IKG, along with the increasing concentration of endosulfan and the length of time of exposure.
