| Abstrak (Bhs. Indonesia) | Periodontitis merupakan penyakit inflamasi kronik pada jaringan periodontal ditandai dengan adanya kehilangan perlekatan dan kehilangan tulang alveolar. Matriks Metaloproteinase-13 (MMP-13) merupakan kolagenase yang disintesis oleh fibroblas berperan dalam mendegradasi matriks ekstraseluler dan mempercepat proses kehilangan tulang. Doxycycline hyclate merupakan antibiotik yang digunakan pada dosis pemeliharaan untuk meningkatkan Tissue Inhibitor Matrix Metalloproteinase (TIMP) pada kondisi periodontitis. Glutathione (GSH) mampu menghambat sintesis MMP-13 oleh fibroblas. Tujuan penelitian ini adalah mengetahui efek terapi kombinasi doxycycline hyclate dan GSH terhadap kadar MMP-13 pada tikus model periodontitis kronis. Jenis penelitian ini adalah eksperimental laboratoris dengan rancangan penelitian posttest only control group design. Penelitian ini menggunakan 35 ekor tikus Sprague Dawley yang dibagi menjadi 5 kelompok: K1 (periodontitis dengan pemberian kombinasi doxycycline hyclate dan GSH), K2 (periodontitis dengan pemberian doxycycline hyclate), K3 (periodontitis dengan pemberian GSH), K4 (kontrol negatif) dan K5 (kontrol sehat). Kadar MMP-13 tikus diperiksa menggunakan uji ELISA didapatkan hasil rerata kadar MMP-13 pada K1 (7,365 ng/ml), K2 (8,495 ng/ml), K3 (9,911 ng/ml), K4 (10,350 ng/ml), K5 (6,504 ng/ml). Hasil penelitian dianalisis menggunakan One Way ANOVA diketahui terdapat perbedaan antar kelompok (p=0,000) dan dilanjutkan dengan post hoc LSD. Hasil uji Pos Hoc diketahui antar kelompok perlakuan (K1, K2, dan K3) dan antara kelompok K4 dan K5 terdapat perbedaan signifikan. Hasil juga menunjukkan tidak terdapat perbedaan antara kelompok K1 dengan K5, dan antara kelompok K3 dengan K4. Hal ini berarti terapi kombinasi Doxycycline hyclate dan GSH memiliki pengaruh positif dalam menurunkan kadar MMP-13 pada perawatan periodontitis kronis. |
| Abtrak (Bhs. Inggris) | Periodontitis is a chronic inflammatory disease in periodontal tissue characterized by attachment loss and alveolar bone loss, which can lead to tooth loss in later stages. Matrix Metaloproteinase-13 (MMP-13) is a collagenase synthesized by fibroblasts that play a role in degrading the extracellular matrix and accelerating the bone loss process. Doxycycline hyclate is an antibiotic that at maintenance doses is used to increase Tissue Inhibitor Matrix Metalloproteinase (TIMP) in periodontitis conditions. Glutathione (GSH) capable of inhibiting MMP-13 synthesis by fibroblasts. The aim of this research was to determine the effect of combination therapy of doxycycline hyclate and GSH on MMP-13 level in chronic periodontitis model of rat. This type of research was experimental laboratory with posttest only control group design. This study used 35 Sprague Dawley rats divided into 5 groups: K1 (periodontitis with combination of doxycycline hyclate and GSH), K2 (periodontitis with doxycycline hyclate), K3 (periodontitis with GSH), K4 (negative control) and K5 (Healthy control). Levels of MMP-13 in rats were examined using ELISA assays showed that the mean of MMP-13 levels on K1 (7,365 ng/ml), K2 (8.495 ng/ml), K3 (9,911 ng/ml), K4 (10,350 ng/ml), and K5 (6,504 ng/ml). The results were analyzed using One Way ANOVA, which known there were differences between groups (p = 0,000) and followed by post hoc LSD. The result of Pos Hoc test was known between treatment group (K1, K2, and K3) and between K4 and K5, there was significant difference with p <0,05. The results also showed no difference between treatment group K1 and K5, and between treatment group K3 and K4, was shown with p> 0,05. This means that combination therapy of Doxycycline hyclate and GSH had positive effect on decreasing MMP-13 in the treatment of chronic periodontitis |
