| Judul Artikel | HUBUNGAN POLIMORFISME GEN CAT TERHADAP KADAR FERRITIN PADA PASIEN THALASSEMIA DI BANYUMAS |
| Abstrak (Bhs. Indonesia) | Latar Belakang: Thalassemia merupakan penyakit autosomal resesif yang paling banyak ditemukan saat ini. Hingga saat ini penatalaksanaan thalassemia yang paling utama adalah transfusi darah. Transfusi darah dan eritropoiesis inefektif menyebabkan iron overload. Pengukuran iron overload dengan menggunakan kadar ferritin. Iron overload meningkatkan produksi ROS dan antioksidan seperti katalase. Polimorfisme gen CAT menyebabkan berkurangnya kemampuan katalase untuk menetralisir H2O2 yang banyak terbentuk akibat peroksidasi lipid. Tujuan: Mengetahui hubungan polimorfisme gen CAT terhadap kadar ferritin pada pasien thalassemia di Banyumas. Desain Penelitian: Penelitian ini merupakan penelitian observasional analitik dengan desain studi cross-sectional. Subjek penelitian adalah pasien thalassemia di RS Banyumas menggunakan teknik random sampling. Didapatkan 58 subjek penelitian yang memenuhi kriteria insklusi dan eksklusi. Kadar ferritin didapatkan dari rekam medis. Polimorfisme gen CAT didapatkan dari hasil PCR-RFLP. Data univariat disajikan dalam bentuk tabel distribusi frekuensi, rerata, dan standar deviasi. Hubungan polimorfisme genotipe gen CAT dengan dengan kadar ferritin dianalisis menggunakan uji one way ANOVA. Sedangkan polimorfisme alel gen CAT dengan kadar ferritin dianalisis menggunakan uji T tidak berpasangan. Hasil: Uji one way ANOVA hubungan antara genotipe polimorfisme gen CAT dengan kadar ferritin diperoleh nilai p=0,255. Uji T tidak berpasangan hubungan antara alel polimorfisme gen CAT terhadap kadar ferritin diperoleh nilai p=0,054. Kesimpulan: Tidak ada hubungan antara polimorfisme gen CAT dengan kadar ferritin pada pasien thalassemia di Banyumas. |
| Abtrak (Bhs. Inggris) | Background: Thalassemia is an autosomal recessive disease most commonly found today. Until now the most important treatment of thalassemia is a blood transfusion. Blood transfusions and ineffective erythropoiesis cause iron overload. Measurement of iron overload by using ferritin levels. Iron overload increases the production of ROS and antioxidants such as catalase. CAT gene polymorphism leads to reduced ability of catalase to neutralize H2O2 that many formed by lipid peroxidation. Objective: To examine the relationship CAT gene polymorphisms and ferritin levels in patients with thalassemia in Banyumas. Study Design: This study is observational analytic with cross-sectional study design. Subjects were patients with thalassemia in RS Banyumas using random sampling techniques. Obtained 58 research subjects who meet the criteria inclusion and exclusion. Ferritin levels obtained from medical records. CAT gene polymorphism results obtained from the PCR-RFLP. The univariat data is presented in the form of frequency, mean, and standard deviation. Relations CAT gene polymorphism genotype with ferritin levels were analyzed using one way ANOVA test. While the CAT gene polymorphism allele with ferritin levels were analyzed using an unpaired t test. Results: One-way ANOVA test CAT gene polymorphism genotype correlation with ferritin levels obtained value of p = 0.255. Unpaired T test CAT gene polymorphism allele relation to the levels of ferritin obtained value of p = 0.054. Conclusion: There is no relationship between CAT gene polymorphism with ferritin levels in patients with thalassemia in Banyumas. |
