Abstrak (Bhs. Indonesia)Daun Kersen (Muntingia calabura L.) memiliki potensi sebagai antiinflamasi dan antioksidan karena mengandung beberapa senyawa aktif berupa flavonoid, saponin, dan tannin. Aktivitas antioksidan dalam daun kersen diketahui dapat menghambat stress oksidatif yang mampu memengaruhi kadar nitrit oksida pada kondisi hiperkolesterolemia. Penelitian ini bertujuan untuk menganalisis pengaruh pemberian ekstrak etanol daun kersen (Muntingia calabura l.) terhadap kadar nitrit oksida tikus putih (Rattus norvegicus) model hiperkolesterolemia. Penelitian ini merupakan penelitian eksperimental dengan post-test only with control group design. Tiga puluh tikus putih jantan galur wistar dibagi dalam 5 kelompok secara acak. Kelompok 1 (kontrol sehat), kelompok 2 (kontrol negatif), kelompok 3 (42 mg/200gramBB), kelompok 4 (84 mg/200gramBB), dan kelompok 5 (168 mg/200gramBB). Induksi hiperkolesterolemia menggunakan minyak babi 2 ml/200gramBB/hari, kuning telur bebek 2 ml/200gramBB/hari, dan pakan buatan berbahan dasar otak sapi 10 gram/100gramBB/tikus. Dilanjutkan pemberian ekstrak etanol daun kersen selama 14 hari. Kadar nitrit oksida diperiksa dengan metode Griess. Rerata kadar NO pada kelompok 1 7,60±0,65 µmol/L, kelompok 2 3,60±0.42 µmol/L, kelompok 3 5,20±0,57 µmol/L, kelompok 4 6,40±0,65 µmol/L, kelompok 5 8,67±0.52 µmol/L. Uji One Way ANOVA kadar NO menunjukkan nilai p=0,000 (p<0,05). Uji post hoc Tukey HSD didapatkan hasil perbedaan rerata yang bermakna diantara semua kelompok data (p<0,05). Pemberian ekstrak etanol daun kersen (Muntingia calabura l.) dapat meningkatkan kadar NO tikus model hiperkolesterolemia, dengan dosis efektif minimum 42 mg/200gramBB.
Abtrak (Bhs. Inggris)Muntingia calabura L. potentially has an effect as an anti-inflammatory and antioxidant from its several active compunds such as flavonoids, saponins, and tannins. The antioxidant activity of Muntingia calabura L. leaves are known to inhibit oxidative stress that can affect the level of nitric oxide in hypercholesterolemic condition. The aim of this study was to analyze the effect of administration Muntingia calabura L. leaves ethanol extract on hypercholesterolemia rat models to NO levels. This was an experimental study using post-test only with control group design. Thirty male wistar rats were randomly assigned to 5 groups. Group 1 as healthy control, group 2 as negative control, group 3, 4, 5, were given ethanol extract of Muntingia calabura L. leaves with the dosage 42mg/200BW, 84 mg/200BW, and 168 mg/200BW. Hypercholesterolemia induction was done using pig oil 2 ml/200gramBW/day, duck eggs yolk 2 ml/200gramBW/day, and modified food made from bovine brain 10 gram/100gramBW/rat. Ethanol extract of Muntingia calabura L. leaves then given for the following 14 days. NO levels were analyzed using Griess test method. Mean value of NO observed on each group was 7,60±0,65 µmol/L for group 1; 3,60±0.42 µmol/L for group 2; 5,20±0,57 µmol/L for group 3; 6,40±0,65 µmol/L for group 4; and 8,67±0.52 µmol/L for group 5. One way ANOVA test of NO level showed the value of p=0,000 (p<0,05). Tukey HSD post hoc test showed a significant mean difference among all groups of the data (p<0,05). Administration of Muntingia calabura L. leaves ethanol extract can increase the levels of NO rat model of hypercholesterolemia, with minimum effective dose of 42 mg/200gramBW.
