| Abstrak (Bhs. Indonesia) | Purwoceng merupakan tanaman endemik yang tumbuh di dataran tinggi Dieng dan banyak dimanfaatkan sebagai obat tradisional Indonesia yang memiliki manfaat afrodisiak. Purwoceng memiliki berbagai zat aktif yang dieksresi melalui ginjal, sehingga berpotensi menimbulkan kerusakan ginjal. Penelitian ini bertujuan untuk menganalisis pengaruh pemberian ekstrak etanol akar Purwoceng (Pimpinella pruatjan Molk.) secara subkronik terhadap kadar ureum dan kreatinin tikus putih (Rattus norvegicus) jantan. Penelitian ini menggunakan metode post test only with control group design. Hewan coba sebanyak 40 tikus dikelompokkan secara acak ke dalam 4 kelompok, yaitu Kelompok A: kontrol (1,5 cc larutan akuades + CMC 1%); Kelompok B: ekstrak etanol akar Purwoceng dengan dosis 42 mg/kgBB/hari + CMC 1%; Kelompok C: ekstrak etanol akar Purwoceng dengan dosis 84 mg/kgBB/hari + CMC 1%; Kelompok D: ekstrak etanol akar Purwoceng dengan dosis 168 mg/kgBB/hari + CMC 1%. Perlakuan diberikan selama 28 hari sesuai panduan uji toksisitas OECD 407. Metode pengukuran kadar ureum menggunakan metode Urease-GLDH (λ 340 nm), dan kreatinin menggunakan metode Jaffe (λ 492 nm). Rerata kadar ureum pada kelompok A 30,40±8,09 mg/dL, kelompok B 36,80±9,05 mg/dL, kelompok C 39,20±7,99 mg/dL dan kelompok D 30,60±5,73 mg/dL. Rerata kadar kreatinin kelompok A 0,57±0,12 mg/dL, kelompok B 0,42±0,09 mg/dL, kelompok C 0,43±0,06 mg/dL, dan kelompok D 0,41±0,04 mg/dL. Uji One-Way ANOVA pada kadar ureum memberikan hasil p=0,034 dan pada kadar kreatinin memberikan hasil p=0,001. Pemberian ekstrak etanol akar purwoceng (Pimpinella pruatjan M.) secara subkronik 28 hari memberikan pengaruh signifikan terhadap peningkatan kadar ureum pada kelompok A dan C, serta penurunan pada kelompok C dan D. Sementara kadar kreatinin tikus putih (Rattus norvegicus) jantan mengalami penurunan signifikan pada kelompok B, C dan D dibandingkan dengan kelompok A. |

| Abtrak (Bhs. Inggris) | Purwoceng is the endemic herb grows in Dieng plateau and has been used as traditional medicine because of its aphrodisiac benefit. Purwoceng has some active chemical compounds excreted through the kidney that potentially damage the kidney. The aim of this study was to analyze the effect of subchronic administration of Purwoceng (Pimpinella pruatjan Molk) roots ethanol extract to the urea and creatinine levels of male albino rat (Rattus norvegicus). The methods of this study was experimental study using post-test only with control group design. Forty male albino rats were randomly divided into four groups. Group A as a control (1,5 cc aquadest + 1% CMC); Group B: 42 mg/kg BW/day Purwoceng roots ethanol extract + 1% CMC; Group C: 84 mg/kg BW/day Purwoceng roots ethanol extract + 1% CMC; and Group D: 168 mg/kg BW/day Purwoceng roots ethanol extract + 1% CMC. Treatment was given during 28 days, with OECD no. 407 guidelines for the procedural reference. Urea level was analyzed using Urease-GLDH method (λ 340 nm), and creatinine level was analyzed using Jaffe method (λ 492 nm). Mean of the Urea levels in group A was 30,40±8,09 mg/dL, group B was 36,80±9,05 mg/dL, group C was 39,20±7,99 mg/dL and group D was 30,60±5,73 mg/dL. Mean of the creatinine levels in group A was 0,57±0,12 mg/dL, group B was 0,42±0,09 mg/dL, group C was 0,43±0,06 mg/dL, and group D was 0,41±0,04 mg/dL. One-Way ANOVA test has p=0,034 value for urea levels and p=0,001 for value creatinine levels. Therefore, subchronic administration of Purwoceng (Pimpinella pruatjan Molk) roots ethanol extract in male albino rats (Rattus norvegicus), could increase urea levels significantly between group A and C, and decrease urea levels significantly between group C and D. Meanwhile, creatinine mean levels decrease significantly between group B, C and D if compared with group A. |
