| Abstrak (Bhs. Indonesia) | Akar purwoceng berkhasiat meningkatkan vitalitas pria. Akar purwoceng mengandung senyawa alkaloid, flavonoid, saponin, dan tanin yang bersifat lipofilik sehingga berpotensi toksik terhadap ginjal. Penelitian ini bertujuan untuk mengetahui efek pemberian ekstrak etanol akar purwoceng secara subkronik terhadap gambaran histologi tubulus proksimal tikus putih (Rattus norvegicus) jantan. Metode penelitian ini adalah eksperimental dengan post-test only with control group design. Empat puluh ekor tikus putih jantan dibagi ke dalam empat kelompok perlakuan secara acak, yaitu kelompok A (kontrol), kelompok B (ekstrak etanol akar purwoceng 42 mg/kgBB), kelompok C (84 mg/kgBB), dan kelompok D (168 mg/kgBB). Perlakuan diberikan selama 28 hari, dan sebelumnya dilakukan aklimatisasi selama 7 hari. Setelah perlakuan usai, hewan coba diterminasi dan diambil ginjalnya untuk diproses menjadi preparat histologi dengan pewarnaan HE. Gambaran histologi ginjal dinilai menggunakan skor nekrosis tubulus proksimal. Rerata skor nekrosis tubulus proksimal paling rendah terdapat pada kelompok A (1,90 ± 0,32). Sedangkan rerata skor kelompok D merupakan rerata paling tinggi (4,50 ± 0,53), diikuti oleh rerata kelompok C (3,50 ± 1,27), dan B (2,20 ± 0,42). Uji Kruskal Wallis menunjukkan nilai p=0,000. Pemberian ekstrak etanol akar purwoceng dosis 42 mg/kgBB, 84 mg/kgBB, dan 168 mg/kgBB secara subkronik menyebabkan kerusakan berupa nekrosis tubulus proksimal tikus putih (Rattus norvegicus) jantan. Dosis toksik subkronik yang mempengaruhi gambaran histologi tubulus proksimal tikus putih (Rattus norvegicus) jantan adalah dosis 84 mg/kgBB/hari. |
| Abtrak (Bhs. Inggris) | Purwoceng roots has the potential effect to increase mans vitality. Purwoceng roots contains alkaloid, flavonoid, saponin, and tanin. These compound are lipophilic and thus potentially toxic to the kidney. The aim of study was to find out the effect of subchronic administration of purwoceng roots ethanol extract to proxsimal tubular histological appearances in male albino rat. This research was an experimental study using post-test only with control group design. Forty rats were randomly divided into 4 groups, that was group A (control group), group B (purwoceng roots ethanol extract of 42 mg/kgBW), group C (84 mg/kgBW), and group D (168 mg/kgBW). All treatmens were given for 28 days, and previously an acclimatization was conducted for 7 days. After the treatment is over, rats were all terminated and the kidneys were took for HE staining. Kidney histological appearances were assessed based on skoring of proximal tubular necrosis. The lowest of mean score of tubular necrosis found in group A (1,90 ± 0,32). The mean score of the group D is the highest (4,50 ± 0,53), followed by mean group of C (3,50 ± 1,27), and B (2,20 ± 0,42). Kruskal-Wallis test showed the value of p = 0,000. The subchronics administration of purwoceng roots ethanol extract with dose 42 mg/kgBW, 84 mg/kgBW, and 168 mg/kgBW caused damage of kidney, that is proxsimal tubular necrosis in male albino rats. The proximal tubular histological appearances was affected by subchronic toxic dose in dose 84 mg/kgBW in mane albino rats. |
