Abstrak (Bhs. Indonesia)Latar Belakang: Karsinoma Nasofaring merupakan tumor ganas yang diakibatkan oleh infeksi virus Epstein-Barr. Infeksi primer terjadi peningkatan titer antibodi IgG dan IgA anti VEB EBNA1 pada serum plasma pasien. Aktivitas mRNA VEB lebih mencerminkan patogenesis KNF dibandingkan diagnosis serologi dan pengukuran DNA VEB di sirkulasi. Pengukuran aktivitas mRNA VEB perlu dilakukan di lokasi tumor primer daerah nasofaring karena gen EBNA1 diekspresikan pada semua daur infeksi VEB. Tujuan: Mengetahui nilai median ekspresi relatif mRNA EBNA1 pada stadium awal dan lanjut penderita KNF. Metode: Rancangan penelitian analitik observasional, studi cross sectional dengan analisis data bivariat. Dilakukan isolasi RNA, sintesis cDNA, dan kuantitas ekspresi mRNA EBNA1 menggunakan RT-qPCR pada 22 sampel tahun 2014. Teknik sampel menggunakan simple random sampling. Menggunakan uji statistik t independen dan penyajian data dengan boxplot. Hasil: Nilai median stadium awal 29.226,71 dan nilai median stadium lanjut 32.429,07. Hasil uji t independen p=0,716. Kesimpulan: Terdapat perbedaan ekspresi relatif mRNA EBNA1 VEB KNF dari biopsi FFPE pada stadium lanjut sebesar 22,67 kali dibandingkan stadium awal. Secara statistik ekspresi relatif mRNA EBNA1 tidak berpotensi sebagai petanda biologi molekul patogenesis KNF, khususnya progresivitas tumor pada stadium lanjut KNF.
Abtrak (Bhs. Inggris)Background: Nasopharyngeal carcinoma is a malignant tumor that caused by the infection of Epstein-Barr virus. In the primary stage of infection the IgG antibody and IgA anti EBV EBNA1 titer of the suspect will increase. The activity of mRNA EBV is more representatif as a pathogenesis marker than serological diagnosis and the EBV DNA measuring in the cirulation. The measuring of mRNA EBV activity needs to be done from the location of the primary tumor in the nasopharynx because this gen is expressed in every cycle of EBV infection. Goals: To figure out the value of median relatif expression of mRNA EBNA1 from early stage and late stage of nasopharyngeal carcinoma patient. Methods: Observasional analytic with crossectional approach and bivariat analysis. RNA isolation, cDNA sinthesis, and mRNA EBNA1 quantity expression were done by RT-qPCR from 22 sample of 2014. With simple random sampling method for the sampling. T test independen statistical method was perfomed and boxplot was used for the data presentation. Result: Median value of early stage cancer was 29.226,71 and for the late stage was 32.429,07. T test independen method showed p=0,716. Conclusion: there was difference of relatif expression of mRNA EBNA1 EBV of nasopharyngeal carcinoma from FFPE biopsy in the late stage diagnosis by 22,67 times than the early stage nasopharyngeal cancer. Statistical method show that relatif expression of mRNA EBNA1 was not potential as a moluclar biology marker for the pathogensis of nasopharyngeal carcinoma, especially for the progressivity of the late stage tumor.
