ABSTRAK

Hiperkolesterolemia pada tikus akibat induksi minyak babi dan PTU (propiltiourasil) menimbulkan peningkatan kadar kolesterol total, trigliserida, LDL (Low Density Lipoprotein), dan penurunan HDL (High Density Lipoprotein). Hiperkolesterolemia menyebabkan perlemakan hepar kemudian menginduksi peroksidasi lipid dan menyebabkan kerusakan hepatosit sehingga mengurangi fungsi sintesis protein. Hiperkolesterolemia juga dapat meningkatkan radikal bebas yang menyebabkan kerusakan jaringan, crosslinking dan fragmentasi DNA dan RNA, mempercepat proteolisis sehingga menurunkan kadar protein total. Simvastatin dan sari markisa ungu memiliki mekanisme yang sama yaitu menghambat HMG-KOA reduktase (Hidroksil Metilglutaril Koenzim A reduktase) sebagai antihiperkolesterolemia. Penelitian ini bertujuan untuk membandingkan efektivitas sari markisa ungu dan simvastatin terhadap kadar protein total pada Rattus norvegicus model hiperkolesterolemia. Metode penelitian yang digunakan adalah eksperimental dengan rancangan post test only with control group. Tiga puluh dua ekor tikus putih jantan dibagi dalam empat kelompok. Kelompok I sebagai kontrol sehat diberi akuades. Kelompok II sebagai kontrol negatif diberi minyak babi 3 mL/200gramBB dua kali sehari dan propiltiourasil 12,5gram dibagi dua kali sehari selama 14 hari. Kelompok III sebagai kelompok perlakuan diberi minyak babi 3 mL/200gramBB dua kali sehari, propiltiourasil 12,5gram dibagi dua kali sehari selama 14 hari, dan sari markisa ungu 4,2 mL/200gramBB/hari selama 14 hari. Kelompok IV sebagai kelompok kontrol positif diberi minyak babi 3 mL/200gramBB dua kali sehari, propiltiourasil 12,5gram dibagi dua kali sehari selama 14 hari, dan simvastatin 0,18mg/kgBB/hari selama 14 hari. Rerata kadar protein total keempat kelompok I, II, III, dan IV adalah 9,00±1,67; 6,17±0,98; 10,50±1,39; dan 9,00±1,55. Hasil uji Kruskal-Wallis yaitu paling tidak terdapat perbedaan kadar protein total yang signifikan antar kelompok (p=0,002) dan hasil uji Post hoc pada kelompok III dan IV yaitu tidak berbeda signifikan (p=0,138). Sari markisa ungu 4,2 mL/200gramBB/hari mempunyai efektivitas yang sama dengan simvastatin 0,18 mg/200gramBB/hari terhadap kadar protein total Rattus norvegicus model hiperkolesterolemia.

ABSTRACT

Hypercholesterolemia in Rattus norvegicus induced pig fat oil and PTU (propylthiouracil) cause increased levels of total cholesterol, triglycerides, LDL (Low Density Lipoprotein), and a decrease in HDL (High Density Lipoprotein). Hypercholesterolemia cause fatty liver induces lipid peroxidation and then cause damage to the hepatocyte thus decreasing the function of protein synthesis. Hypercholesterolemia can also increase free radicals that cause tissue damage, crosslinking and fragmentation of DNA and RNA, accelerate proteolysis resulting in lower total protein levels. Simvastatin and Passiflora edulis var edulis juice have the same mechanism that inhibits HMG-COA reductase (Hydroxyl Methylglutaryl Coenzyme A Reductase) as inhibits hypercholesterolemia. This study aimed to compare the effectiveness of Passiflora edulis var edulis juice and simvastatin on levels of total protein in rats (Rattus norvegicus) model of hypercholesterolemia. The design method was experimental with post test only control group design. Thirty two male rats were divided into four groups. Group I as normal control group was given aquades. Group II as a negative control by pig fat oil 3mL/200gBW rat twice a day and propylthiouracil 12,5gram divided twice daily for 14 days. Group III as the treatment group were given pig fat oil 3 mL/200 gBW rat, propylthiouracil 12,5gram divided twice daily for 14 days, and Passiflora edulis var edulis juice 4.2mL/200gBW rat/day for 14 days. Group IV as the positive control group was given pig fat oil 3mL/200 gBW rat/day, propylthiouracil 12,5gram divided twice daily for 14 days, and simvastatin 0.18mg/200 gBW/day for 14 days. The mean of total protein level in four groups I, II, III, and IV are 9.00 ± 1.67; 6.17 ± 0.98; 10.50 ± 1.39; and 9.00 ± 1.55. Kruskal-Wallis test results that at least there is no difference in total protein levels were significantly between groups (p = 0.002) and Post hoc test results in group III and IV are not significantly different (p = 0.138). Passiflora edulis var edulis juice 4.2 mL/200gBW /day has the same effectiveness with simvastatin 0.18 mg/200gBW/day of the total protein levels Rattus norvegicus model hypercholesterolemia.
