Abstrak (Bhs. Indonesia)INH mempunyai peranan dalam menghambat sintesis asam mikolat yang merupakan komponen pembentuk dinding sel M. tuberculosis. Mengetahui pengaruh pemaparan obat INH terhadap karakteristik M. tuberculosis dan terjadinya resistensi INH. Metode penelitian eksperimen dengan menggunakan Rancangan percobaan acak Lengkap dengan variasi waktu pemaparan 24 jam, 48 jam, 72 jam dan 96 jam pada konsentrasi INH 0,5 μg/ml dengan jumlah ulangan 6. Hasil penelitian menunjukkan ada pengaruh pada masing-masing perlakuan INH 0,5 μg/ml terhadap karakteristik M. tuberculosis, pada pemaparan 24 jam diperoleh persentase pertumbuhan 18,25%, ukuran rerata sel 1,64 x 0,35 µm, persentase kerusakan sel 28,1%, rerata pertumbuhan pada NaCl 5% sebanyak 1 koloni, uji katalase semi kuantitatif 3,6 mm. Pada pemaparan 48 jam diperoleh persentase pertumbuhan sel 7,9%, ukuran rerata sel 1,64 x 0,36 µm, persentase kerusakan sel 41%, rerata pertumbuhan pada NaCl 5% sebanyak 4 koloni, uji katalase semi kuantitatif 5,4 mm. Pada pemaparan 72 jam diperoleh persentase pertumbuhan sel 7,0%, ukuran rerata sel 1,64 x 0,32 µm, persentase kerusakan sel 53,4%, rerata pertumbuhan pada NaCl 5% tidak ada pertumbuhan, uji katalase semi kuantitatif 3,6 mm, pada pemaparan 96 jam diperoleh persentase pertumbuhan sel 1,6%, ukuran rerata sel 1,61 x 0,29 µm, persentase kerusakan sel 57%, rerata pertumbuhan pada NaCl 5% sebanyak 3 koloni, uji katalase semi kuantitatif 15 mm. Perubahan morfologi sel yang terpapar INH 0,5 μg/ml pada permukaan sel terjadi kerutan dan penyusutan panjang sel setelah regenerasi, permukaan sel mulai kasar dan sedikit ada kerusakan, sel mengalami lisis dan sebagian sel berbentuk oval , sel mengalami perubahan bentuk menjadi kokus. Uji drung susceptibility tes (DST) pada pemaparan INH 0,5 μg/ml selama 24 jam, 48 jam, 72 jam hasil sensitif, pada pemaparan 96 jam hasil resiten.karakteristik M. tuberculosis yang terpapar INH 0,5 µg/ml mengalami perubahan morfologi sel, menjadi lebih pendek dan perubahan bentuk dari batang menjadi kokus, namun perubahan morfologi ini tidak terjadi secara permanen pada generasi berikutnya. Berdasarkan uji statistik pertumbuhan M. tuberculosis yang terpapar dengan INH 0,5 µg/ml pada masing-masing perlakuan mempunyai pengaruh yang sangat signifikan pada kadar NaCl 5% dan tes katalase semi kuantitatif. Uji DST M. tuberculosis yang terpapar INH 0,5 µg/ml selama 96 jam mengalami resistensi sebesar 2% dari 108 CFU/ml.
Abtrak (Bhs. Inggris)INH’s role in inhibiting the synthesis of mycolic acid which is a component of the cell wall of M. tuberculosis. Research objectives to determine the effect of INH drug exposure on the characteristics of M. tuberculosis and INH resistance occurrence. The experiment research method using a complete randomized experimental design with variations in exposure time 24 hours, 48 hours, 72 hours and 96 hours on INH concentration of 0.5 μg/ ml with 6 replications. The results showed influence on each 0.5 μg/ml INH treatment towards the characteristics of M. tuberculosis, at 24 hours of exposure was obtained 18.25% percentage growth, the average cell size was 1.64 x 0.35 μm, the percentage of cell damage was 28.1%, the average growth in 5% NaCl was 1 colony, semi-quantitative catalase test was 3.6 mm. At 48 hours of exposure was obtained 7.9% percentage of cell growth, the average cell size was 1.64 x 0.36 μm, the percentage of cell damage was 41%, the average growth in 5% NaCl were four colonies, semi-quantitative catalase test was 5.4 mm. At 72 hours of exposure was obtained 7.0% percentage of cell growth, the average cell size was 1.64 x 0.32 μm, the percentage of cell damage was 53.4%, no growth had been found in 5% NaCl, semi-quantitative catalase test was 3.6 mm. At 96 hours of exposure was obtained 1.6% percentage of cell growth, the average cell size was 1.61 x 0.29 μm, the percentage of cell damage was 57%, the average growth in 5% NaCl were 3 colonies, semi-quantitative catalase test is 15 mm. Morphological changes in cells exposed to 0.5 μg/ml INH were wrinkles on the cell surface and length shrinkage occur after the regeneration of cells, the cells surface began to become rough and has little damage, the cells experience lysis and the half part of it was oval, the cell’s shape changes into cocci. Test of drug susceptibility test (DST) on 0.5 μg/ml INH exposure for 24 hours, 48 hours, 72 hours was sensitive results, 96 hours exposure was resistance results. The characteristics of M. tuberculosis exposed to INH 0.5 ug/ml were changes in cell morphology, shorter and change form rods into cocci. However, the morphological changes did not endure permanently in the next generation. Based on statistical test of the growth of M. tuberculosis exposed by 0.5 ug/ml INH in each treatment had a very significant influence on the level of 5% NaCl and semi-quantitative catalase test. M. tuberculosis DST test exposed by 0.5 ug/ml INH for 96 hours experienced resistance by 2% of 108 CFU / ml.
