Abstrak (Bhs. Indonesia)Grouper are important fisheries commodity because it is one of export oriented products. This entails to high economic value. Grouper are widely distributed across Indonesia. It can be found in Sumatra, Java, Sulawesi, and Ambon reef ecosystems. Sulawesi has a potential catchment are e.g. Spermonde Archipelago, South Sulawesi. Leopard grouper (Plectropomus leopardus) is among the economically important species. This species is also exploited in Spermonde Archipelago, including Reang-Reang Island. Spermonde Archipelago’s fisherman collected leopard grouper using destructive methods, such as banning and cyanide fishing. These fishing methods are contradictive with conservation principles. Therefore, various conservation efforts are needed. Systematic study, especially, intra-populational variation study is among of the important starting point. Intra-populational variation data can be obtained from a study using cytochrome c oxidase 1 (COI) gene. This research aims to obtain genetic diversity data on leopard grouper population in Reang-Reang Island based on partial sequences of cytochrome C oxidase 1 gene. The research used survey method. Caudal fin samples were obtain from Animal Taxonomy Laboratory Biology Faculty of Unsoed collection. Grouper DNA is extracted, then performed with the techniques of PCR amplification. The sequence of genetic diversity has on the PCR was then edited using the Bio-Edit and statistically analyzed using analysis of molecular variance (AMOVA) with the help of Arlequin software. Fragments that have been amplificated will be in order to see the sequencing of DNA. Characters can be used as molecular genetic marker is a cytochrome oxidase 1 gene (CO1). The variables observed in this study i.e. intrapopulation genetic diversity. The calculated parameters include gene CO1 polymorphism, haplotype diversity, and nucleotides diversity.
The Arlequin K2P model analysis showed high haplotype diversity with the value of 1.0000 +/- 0.0764 and high nucleotide diversity at 1,5 % +/- 0,9 %. Polymorphism analysis showed polymorphism sites percentage of 5,015198%. This mean that COI gene of leopard grous was polimorphic.
