Abstrak (Bhs. Indonesia)Produksi GH dilaporkan berkorelasi positif dengan pertumbuhan ikan. Ada indikasi variasi pertumbuhan juga dipengaruhi oleh variasi sekuen gen GH. Tujuan dari penelitian ini yaitu untuk mengindentifikasi gen GH dengan teknik RT-PCR dan mengetahui variasi susunan basa nukleotida gen GH pada lele dumbo (Clarias gariepinus) berbeda ukuran dan lele dumbo yang berbeda strain. Sampel RNA diekstraksi dari empat kelompok ikan, dua kelompok berbeda ukuran dari strain yang sama dan dua kelompok berbeda strain berukuran sama. Sampel RNA dari hipofisa ikan digunakan pada teknik RT-PCR untuk amplifikasi gen GH. Produk RT-PCR disekuensing untuk kemudian dianalisis BLAST, analisis multiple sequences alignment (MSA), dan analisis filogenetik. Hasil analisis BLAST menunjukkan bahwa produk RT-PCR terkonfirmasi sebagai bagian gen GH dari lele dumbo. Hasil analisis MSA menunjukkan lele dumbo ukuran berbeda mempunyai sekuen nukleotida gen GH yang sama. Pada sekuen nukleotida variasi gen GH ditemukan pada lele dumbo yang berbeda strainnya. Namun demikian variasi tersebut tidak menyebabkan perbedaan urutan asam amino GH dari semua sampel.
Abtrak (Bhs. Inggris)Production level of growth hormone (GH) was reported to have positive correlation with fish growth rate. There was indication that growth rate variation also was affected by variation of GH gene sequences. Aims of this research were to identify GH gene by RT-PCR technique and to determine variation of nucleotide base sequence of the gene from African catfish with different size and different strain. Sample of RNA was extracted from pituitary gland of four fish group, two group were from different size and same strain and two group were from different strain with similar size. Sample of RNA was used in RT-PCR technique to amplify GH gene. Product of RT-PCR was sequenced to be analyzed by BLAST analysis, multiple sequences alignment (MSA) analysis, and phylogenetic analysis. Result of BLAST analysis showed that RT-PCR product was confirmed as partial fragmen of GH gene of African catfish. Result of MSA analysis showed that different size of African catfish from a strain have same GH gene sequence. Variation of GH gene nucleotide sequence was found from different strain of the fish. However, this variation did not induce different sequence of GH amino acid from all samples.
