Abstrak (Bhs. Indonesia)Kadal (Eutropis multifasciata Kuhl) merupakan salah satu jenis reptilia yang berhabitat di hutan maupun di semak-semak. Fertilisasi E. multifasciata Kuhl bersifat internal dan perkembangan embrionya bersifat ovovivipar, sehingga zigot tumbuh dan berkembang menjadi embrio di dalam uterus. Meskipun demikian oosit memiliki massa yolk yang banyak. Beberapa aspek reproduksi telah diteliti, namun informasi mengenai perkembangan embrionya masih terbatas. Hal tersebut mendorong untuk dilakukannya penelitian mengenai perkembangan embrio E. multifasciata Kuhl. Penelitian ini dilakukan untuk mengevaluasi perkembangan embrio berdasarkan ciri-ciri morfologi dan histologis.
Pengambilan sampel kadal dilakukan secara acak (random sampling) setiap minggu selama periode Oktober-Desember 2015. Kadal betina yang tertangkap dianastesi kemudian dibedah untuk diamati perkembangan embrionya. Embrio diangkat dan dievaluasi tahapan perkembangannya menggunakan mikroskop stereo. Embrio pada tahap pre-neurula dan neurula dievaluasi berdasarkan struktur yang sudah terbentuk melalui pembuatan preparat histologis. Embrio tahap organogenesis dievaluasi dan diidentifikasi berdasarkan struktur morfologi yang sudah terbentuk.
Hasil penelitian menunjukkan bahwa pada periode Oktober 2015 didapatkan 48 embrio, dengan tahapan perkembangan pre-neurula sebesar 33,33%, neurula 11,11% dan organogenesis 55,56%. Pada bulan November 2015 diperoleh 39 embrio dengan tahapan pre-neurula sebesar 28,57% dan organogenesis 71,43%. Pada bulan Desember 2015 diperoleh 51 embrio dengan tahapan pre-neurula sebesar 40% dan organogenesis sebesar 60%. Proses kalsifikasi pada tulang embrio pertama kali terdeteksi dengan pewarnaan Alizarin red pada saat embrio mencapai tahap pembentukan digiti. Puncak perkembangan embrio diduga terjadi pada periode Desember ditandai dengan diperolehnya fetus lengkap (siap lahir). Data tersebut mengindikasikan bahwa periode Oktober-Desember merupakan waktu yang sesuai untuk perkembangan embrio sebagaimana ditunjukkan oleh tingginya persentase embrio pada tahap organogenesis.
Abtrak (Bhs. Inggris)The Javan sun skink (Eutropis multifasciata Kuhl) is a member of Reptile inhabits forest and scrubland. This species is an ovoviviparous animal in which fertilization occurs internally, embryos develop in the female reproductive tracts but the oocytes contain large amount of yolk. Some reproductive aspect have been reported by several researchers yet less information is available on its embryonic development. Therefore this study was conducted to evaluate embryonic development of this species based on morphological or histological features.
The adult Javan sun skink were captured weekly according to random sampling technique during October until December 2015. The females were anesthetized and sacrified to examine the availability of embryos. The embryos and fetuses were removed from the gravid females and closely examined under the stereo microscope. The embryos were categorized into preneurula, neurula and organogenesis stage. The morphological features of preneurula and neurula embryos were recorded then the embryos were processed for paraffin embedded histological sections. The embryos at organogenesis stage were examined based on morphological features.
The results showed that in October 2015 there were 48 embryos, 33.33% of them were preneurula, 11.11% were neurula and 55.56% were at organogenesis stage. In November there were 39 embryos, 28.57% of them were preneurula and 71.43% embryos were at organogenesis stage. In December 2015, there were 51 embryos, 40% them were preneurula and 60% embryos were at organogenesis stage. Bone calcification as indicated by Alizarin red was first to be detected at embryos showing digity formation. The most advance embryonic development was observed in December. These data suggested that October-December was a suitable time for embryonic development this species as indicated by a higher percentage of embryo in organogenesis stage.
