Abstrak (Bhs. Indonesia)Dalam penelitian ini telah dikembangkan metode kolorimetri
menggunakan pencitraan kamera handphone untuk menentukan kadar glukosa
dan validasi metode tersebut. Penentuan glukosa secara kolorimetri didasarkan
pada reaksi glukosa dengan enzim glukosa oksidase yang menghasilkan H2O2
yang kemudian bereaksi dengan KMnO4 sebagai indikator redoks. Perubahan
warna KMnO4 diukur menggunakan kamera handphone dan dianalisis
menggunakan program Color Grab. Validasi metode menunjukkan sensitivitas
tertinggi pada intensitas warna hijau dengan nilai KV 1,682%; HORRAT 0,314;
recovery 95,97%; rentang konsentrasi linear 0,5 mM sampai dengan 3,0 mM
dengan nilai koefisien korelasi 0,9991. Batas deteksi (LOD) 0,13 mM; batas
kuantifikasi (LOQ) 0,45 mM; rentang metode 0,45-38,78 mM. Selektivitas
KMnO4 kurang selektif untuk menentukan H2O2 dengan adanya asam askorbat.
Hasil analisis kadar glukosa dalam sampel menggunakan kamera handphone
tidak berbeda secara signifikan dari hasil analisis menggunakan spektrofotometer
UV-Vis (P>0,05).
Abtrak (Bhs. Inggris)In this research a colorimetric method using a handphone camera
imaging to determine the concentration of glucose and as method validation have
been developed. The colorimetric glucose determination was based on the
reaction of glucose with glucose oxidase enzyme resulted in H2O2 which was
finally reacted with KMnO4 as redox indicators. The color change of KMnO4 was
measured using a handphone camera and analyzed using Color Grab program.
The method validation showed the highest sensitivity of the intensity was the
green color with KV value of 1,682%; HORRAT of 0,314; recovery of 95,97%;
linear range of 0,5 mM to 3,0 mM with a correlation coefficient of 0,9991. The
limit of detection (LOD) was 0,13 mM; limit of quantification (LOQ) was 0,45
mM; methods range of 0,45–38,78 mM. The selectivity of KMnO4 was less
selective to H2O2 determination in the presence of ascorbic acid. The results of
the analysis of glucose levels in the samples using a handphone camera were not
significantly different from the results of the analysis using UV-Vis
spectrophotometer (P>0,05).
