| Abstrak (Bhs. Indonesia) | ABSTRAK Latar Belakang: Gen MLH-1 adalah suatu gen missmatch repair (MMR) pada jalur karsinogenesis karsinoma kolorektal. Ekspresi protein MLH-1 positif pada penderita menunjukkan prognosis yang lebih baik dibandingkan ekspresi negatif. Penelitian mengenai profil ekspresi MLH-1 dan klinikopatologik karsinoma belum pernah dilakukan di wilayah Banyumas, Jawa Tengah, Indonesia. Tujuan: Mengetahui profil klinikopatologik (jenis kelamin, usia, topografi, stadium dukes, dan derajat diferensiasi) dan ekspresi MLH-1 pada karsinoma kolorektal. Metode: Sampel blok parafin yang berasal dari 45 kasus adenokarsinoma kolorektal post-hemikolektomi dikumpulkan sejak Januari hingga Desember 2014 di RSUS Prof. Margono Soekarjo Purwokerto dan Laboratorium Rujukan Banyumas, Jawa Tengah. Ekspresi MLH-1 diteliti melalui immunohistokimia menggunakan antibodi poliklonal (Biocare Medical; USA). Kriteria pewarnaan sel diklasifikasikan melalui metode skoring allred. Karakteristik klinikopatologik yang dikumpulkan meliputi jenis kelamin, usia, topografi, stadium dukes, dan derajat diferensiasi dinilai dari 57 sampel total. Hasil: Pewarnaan MLH-1 menunjukkan hasil positif 20% dan negatif 80%. Karakteristik klinikopatologik menunjukkan jenis kelamin laki-laki (56,1%), perempuan (43,9%), usia ≤40 tahun (19,3%) dan >40 tahun (80,7%), topografi kolon proksimal (40,4%) dan kolon distal (59,6%), stadium Dukes A (0%), B (70,2%), C (17,5%), D (12,3%), derajat diferensiasi baik (84,2%), moderat (14,0%), buruk (1,8%). Kesimpulan: Karakteristik klinikopatologik karsinoma kolorektal di wilayah Banyumas mayoritas berjenis kelamin laki-laki, usia terjadi pada usia >40 tahun, topografi pada kolon kiri, stadium Dukes pada stadium Dukes B, dan derajat diferensiasi baik. |
| Abtrak (Bhs. Inggris) | ABSTRACT Background: MLH-1 is a mismatch repair (MMR) gene which classified as colorectal cancer carcinogenesis pathaway. Patients with positive expression of MLH-1 are known to have better prognosis. There has not any study about expression of MLH-1 and clinicopathological profile of colorectal cancer in Banyumas region, Central of Java, Indonesia. Aim: To discover the expression of MLH1 and clinicopathologic characteristics (including gender, age, site of tumor, Dukes staging, and histopatological grading of colorectal cancer. Method: Paraffin-preserved adenocarcinoma colorectal samples of 45 cases were collected from patients undergoing hemicolectomy from January through December 2014 in Prof. Margono Soekarjo Purwokerto Hospital and Banyumas referral laboratory, Central of Java. MLH-1 expressions were studied using polyclonal antibody (Biocare Medical:USA). The number of cells that immunohistochemistry stained were assesed using allred score methods. Clinicopathologic data that assesed from 57 total sampling including gender, age, tumor location, Dukes staging and histopathological grading. Result: MLH-1 expression which showed positive and negative staining were 20% and 80%.Clinicopathological characteristic assesment results were male gender (56,1%), female (43,9%), ≤40 years of age (19,3%), >40 years (80,7%), distal colon sites of tumor (40,4%), proxymal colon (59,6%), Dukes staging A (0%), B (70,2%), C (17,5%), D (12,3%), and well-differentiated histopathological grading (84,2%), moderate-differentiated (14,0%), poor-differentiated (1,8%). Conclusion: The majority both clinicopathologic characteristic and MLH-1 expression of colorectal cancer in Banyumas region were male gender, >40 years of age, right sided colon sites of tumor, Dukes staging B, and well-differentiated histopathological grading. |
