| Abstrak (Bhs. Indonesia) | Latar Belakang: Ekspresi COX-2 berperan penting pada proses inflamasi. Kunyit diharapkan dapat menghambat ekspresi COX-2 sehingga risiko kerusakan akibat inflamasi dapat dikurangi. Tujuan: Mengetahui efek anti-inflamasi ekstrak etanol kunyit (Curcuma domestica) terhadap ekspresi COX-2 pada sel RAW 264.7 yang diinduksi LPS pada dosis 62,5 µg/ml, 125 µg/ml, dan 250 µg/ml. Metode: Penelitian ini merupakan penelitian ekperimental post-test only with control group design. Sel RAW 264.7 dibiakan di dalam media kultur Dublecco’s Modified Eagle Medium (DMEM). Biakan sel dibagi menjadi 6 kelompok (K) uji pada 24-well plate; KA (kontrol tanpa LPS), KB (Kontrol LPS), KC (kontrol tanpa antibodi primer), KD (perlakuan 62,5μg/mL), KE (perlakuan 125μg/mL), dan KF (perlakuan 250μg/mL). Setelah 18 jam perlakuan semua sel difiksasi menggunakan etanol bertingkat kemudian dilakukan uji imunositokimia. Hasil: Jumlah ekspresi COX-2 KA (10,515,15), KB (82,29±1,49), KC (29,01±5,19), KD (32,19±5,36), KE (24,29±5,88), dan KF (40,42±3,15). Hasil uji one way ANOVA menunjukkan perbedaan yang signifikan (p<0,05), uji Post Hoc Tamhanes menunjukan perbedaan signifikan (p<0,05) dengan KE menunjukan perbedaan rerata paling besar dari KB dibandingkan kelompok perlakuan lain (57,99). Kesimpulan: Terdapat efek anti-inflamasi ekstrak ekstrak etanol kunyit (curcuma domestica) terhadap ekspresi COX-2 pada sel RAW 264,7 yang diinduksi lipopolisakarida dengan dosis optimal ekstrak etanol kunyit pada 125μg/mL. |
| Abtrak (Bhs. Inggris) | Background: Expression of COX-2 has the important role in inflammation process. Turmeric extract is expected can inhibit COX-2 and the damage caused by inflammation can be reduced. Objective: This study was conducted to know anti-inflammatory effect of turmeric (Curcuma domestica) ethanol extract against COX-2 expression in RAW 264.7 cell-induced by lipopolysaccharide using 62,5 µg/ml, 125 µg/ml, dan 250 µg/ml doses. Method: This study was an experimental post-test only with control group study design. RAW 264.7 cell is cultured in Dublecco’s Modified Eagle Medium (DMEM) culture medium. This culture medium contain 1% penicilin-streptomycin and 10% fetal bovine serum (FBS). Cultured cell was distributed into six groups (G) at 24-well plate; AG (control without LPS), BG (control with LPS), CG (control without primary antibody), DG (LPS with 62,5μg/mL of turmeric extract as treatment), EG (treatment 125μg/mL), and FG (treatment 250μg/mL). Eighteen hours after administration each group were being fixated by graded series of ethanol and prepared for immunocytochemistry. Result: COX-2 expression of AG (10,515,15), BG (82,29±1,49), CG (29,01±5,19), DG (32,19±5,36), EG (24,29±5,88), and FG (40,42±3,15). Statistical analysis with one way ANOVA reveal significant difference (p<0,05). Post Hoc Tamhane’s shown significant difference (p<0,05). EG has the most significant mean difference from BG compared with another treatment group (57,99). Conclusion: There was anti-inflammatory effect of turmeric (Curcuma domestica) ethanol extract against COX-2 expression in RAW 264.7 cell-induced lipopolysaccharide. Turmeric extract has the lowest COX-2 expression at 125μg/mL. |
