Bunga kecombrang (Nicolaia speciosa) dengan kandungan kimia flavonoid, alkaloid, steroid, dan triterpenoid memiliki aktivitas antibakteri. Tujuan penelitian ini adalah untuk membuat formula gel ekstrak bunga kecombrang dengan basis carbopol 940 yang memenuhi syarat evaluasi selama penyimpanan, serta mengetahui aktivitas antibakterinya terhadap Staphylococcus aureus.
Ekstraksi simplisia bunga kecombrang dilakukan secara maserasi bertingkat menggunakan pelarut n-heksan dan etil asetat. Formula gel dibuat dengan variasi kadar carbopol 940 (0,5%, 1%, 1,5%, dan 2% b/v) mengandung 3% b/v ekstrak bunga kecombrang. Formula terbaik yang memenuhi syarat sifat fisik dan stabilitas sediaan diuji aktivitas antibakterinya terhadap Staphylococcus aureus menggunakan metode difusi sumuran. Data organoleptik, homogenitas dan pH dianalisis secara deskriptif, sedangkan data viskositas, daya sebar, dan daya lekat dianalisis menggunakan Anova satu arah dilanjutkan dengan uji LSD/Dunnet T3. Data zona hambat bakteri formula terbaik dibandingkan dengan kontrol positif (gel Clindamycin) dan negatif menggunakan uji T independen.
Hasil penelitian menunjukkan bahwa kadar Carbopol 940 dapat mempengaruhi sifat fisik dan stabilitas sediaan. Formula 1 merupakan formula terbaik yang memenuhi syarat sifat fisik dan stabilitas sediaan dengan nilai pH 6, viskositas 226.800-396.000 cps, daya lekat 1,00-1,92 detik, dan daya sebar 5,0-5,2 cm. Formula 1 menunjukkan aktivitas antibakteri dibanding kontrol negatif dengan zona hambat sebesar 4,23 mm dan tidak berbeda signifikan dengan kontrol positif gel Clindamycin. Namun viskositas Formula 1 sangat tinggi dan aktivitas antibakterinya lebih rendah dibanding ekstrak tanpa formulasi. Sehingga perlu dilakukan formulasi bentuk sediaan topikal lain untuk mengoptimalkan sifat fisik dan aktivitas antibakteri sediaan bunga kecombrang.

Etlingera flower (Nicolaia speciosa) which contains chemical compounds such as flavonoids, alkaloid, steroids and triterpenoids have an activity as antibacterial agents. The purpose of this research is to make a gel formulation of etlingera flower extract with Carbopol 940 that qualified from required of formulation evaluation during storage, and to know the activity of antibacterial agents againts Staphylococcus aureus.
Extraction of etlingera flower’s simplisia is carried out by storied maceration using n-hexane and ethyl acetate solvents. A gel formulation are made with variation of Carbopol 940 (0,5%, 1%, 1,5% and 2% b/v) contains 3% b/v etlingera flower’s extract. The best formula which qualified from evaluation of gel formulation is tested for an antibacterial activity against Staphylococcus aureus using well diffusion method. The organoleptyc data, homogenity and pH were analyzed descriptively, while the viscosity data, dispersive power, and the adhesive power was analyzed using one way ANOVA continued with LSD/Dunnet T3 test. The inhibition zone’s best formula data is compared with the positive (Clindamycin gel) and negative control using an independent T test.
The results showed that levels of Carbopol 940 may affect the physical properties and stability of the preparations. Formula 1 is the best formula qualified physical properties and stability of the preparation with a pH value of 6, 226.800 to 396.000 cps of viscosity, adhesive power 1,00 to 1,92 seconds, and the dispersive power from 5,0 to 5,2 cm. Formula 1 shows the antibacterial activity compared negative control with inhibition zone of 4,23 mm and do not differ significantly from the positive control of Clindamycin gel. But Formula 1’s viscosity is very high and antibacterial activity is lower than the extract without formulations. So we need another topical formulation dosage forms to optimize the physical properties and the antibacterial activity of etlingera flower’s preparations.
