| Abstrak (Bhs. Indonesia) | Merkuri adalah polutan yang sangat beracun dan persisten yang dapat mencemari perairan sungai. Dampak HgCl2 bagi biota perairan menghambat reproduksi terutama ekspresi gen cGnRH-II. Biota uji yang digunakan yaitu ikan nilem. Tujuan penelitian ini yaitu Mengetahui ekspresi gen cGnRH-II yang dipengaruhi oleh berbagai konsentrasi merkuri terhadap ikan nilem (Osteochilus hasselti), mengetahui konsentrasi merkuri yang mengganggu ekspresi gen cGnRH-II pada ikan nilem (Osteochilus hasselti) dan mengetahui korelasi antara merkuri dengan ekspresi gen cGnRH-II, merkuri dengan IGS, dan ekspresi gen cGnRH-II dengan IGS. Metode yang digunakan adalah metode eksperimental, dengan desain penelitian Rancangan Acak Lengkap (RAL). Penelitian dilakukan dengan 4 tahap, pengambilan hipotalamus, isolasi RNA, DNAse Treatment, Pengukuran konsentrasi RNA dan Real-Time PCR. Sampel diberi 4 perlakuan logam berat berbeda selama 60 hari dengan pengambilan sampel setiap 2 minggu sekali. Data kuantitatif berupa ekspresi gen penghasil GnRH. Hasil penelitian menunjukkan bahwa pengaruh perlakuan merkuri selama 60 hari terhadap nilai ekspresi gen cGnRH-II memiliki kisaran rata-rata 0,53-2,36. Pengaruh perlakuan merkuri berupa penurunan nilai konsentrasi ekspresi gen cGnRH-II dan nilai IGS, seiring dengan meningkatnya konsentrasi merkuri serta lamanya waktu pemaparan. | Abtrak (Bhs. Inggris) | Mercury is a highly toxic pollutant and persistent that could contaminate the river waters. The Impact of HgCl2 for aquatic biota inhibit reproduction, especially gene expression cGnRH-II. Biota test used in this research is fish nilem. The purpose of the research are to know the gene expression cGnRH-II influenced by various concentrations of mercury to nilem fish (Osteochilus hasselti), to determine the concentration of mercury disrupt the gene expression cGnRH-II in nilem fish (Osteochilus hasselti) and to determine the correlation between the heavy metal mercury with expression cGnRH-II genes, mercury with IGS, and gene expression cGnRH-II with IGS. The method used was experimental method, the research design completely randomized design (CRD). The study was conducted with 4 stages, making the hypothalamus, RNA isolation, DNAse treatment, RNA concentration measurements and Real-Time PCR. Samples were given 4 different treatment of heavy metal for 60 days with sampling every 2 weeks. The quantitative data in the form of GnRH producing gene expression. The results showed that the treatment effect of mercury for 60 days against the concentration of gene expression cGnRH-II has average range 0.53 to 2.36. HgCl2 treatment effect of mercury in the form of a decrease in the concentration of gene expression IGS cGnRH-II and value, along with the increasing mercury concentrations and exposure times.
