| Abstrak (Bhs. Indonesia) | Limbah industri sebagai pencemar antropogenik yang mengandung logam berat. HgCl2 adalah polutan yang sangat beracun dan persisten yang dapat mencemari perairan sungai. Dampak HgCl2 bagi biota perairan menghambat reproduksi terutama ekspresi gen sGnRH. Biota uji yang digunakan yaitu ikan nilem. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh logam berat merkuri klorida (HgCl2) terhadap ekspresi gen sGnRH serta konsentrasi logam berat HgCl2 yang dapat mengganggu ekspresi gen sGnRH pada ikan nilem (Osteochilus hasselti) betina. Metode yang digunakan adalah metode eksperimental, dengan desain penelitian Rancangan Acak Lengkap (RAL). Penelitian dilakukan dengan 4 tahap, pengambilan hipotalamus, isolasi RNA, DNAse Treatment, Pengukuran konsentrasi RNA dan Real-Time PCR. Sampel diberi 4 perlakuan logam berat berbeda selama 60 hari dengan pengambilan sampel setiap 2 minggu sekali. Data kuantitatif berupa ekspresi gen penghasil GnRH dianalisis dengan one ways Anova dan Uji BNT. Hasil penelitian menunjukkan bahwa pengaruh perlakuan logam berat HgCl2 selama 60 hari terhadap nilai konsentrasi ekspresi gen sGnRH memiliki kisaran rata-rata 0,90-42,97. Pengaruh perlakuan logam berat HgCl2 berupa penurunan nilai konsentrasi ekspresi gen sGnRH dan nilai IKG, seiring dengan meningkatnya konsentrasi HgCl2 serta lamanya waktu pemaparan.

The Impact of HgCl2 for aquatic organism primarily to lower reproductive such as sGnRH gene expression. Osteochilus hasselti is object for gene expression test. The purpose of this study was to determine the effect of mercury chloride (HgCl2) on gene expression sGnRH and concentrations of (HgCl2) that can interfere with sGnRH gene expression in females Osteochilus hasselti fish. The method used was experimental method, the research design Completely Randomized Design. The study was conducted with 4 stages, making the hypothalamus, RNA isolation, DNAse treatment, RNA concentration measurements and Real-Time PCR. Samples were given four different treatment of heavy metal for 60 days with sampling every 2 weeks. The quantitative data in the form of GnRH-producing gene expression was analyzed by one-ways ANOVA and BNT test. The results showed that the treatment effect of heavy metals HgCl2 for 60 days against the value of the concentration of gene expression sGnRH have an average range of 0.90 to 42.97. HgCl2 treatment effect of heavy metals in the form of a decrease in the concentration of sGnRH gene expression and IKG value, along with the increasing concentration of HgCl2 and the length of time of exposure. |
