| Abstrak (Bhs. Indonesia) | Ashitaba (Angelica keiskei) merupakan tanaman obat tradisional dari Jepang. Ekstrak daun ashitaba mengandung senyawa bioaktif golongan alkaloid, flavonoid, tanin, saponin, terpenoid dan fenol yang berpotensi sebagai antibakteri. Penelitian ini dilakukan untuk mengetahui pengaruh ekstrak etanol daun ashitaba terhadap atkitivas pertumbuhan bakteri patogen Gram positif dan Gram negatif, mengetahui perbedaan zona hambat diantara variasi konsentrasi ekstrak terhadap pertumbuhan bakteri patogen Gram positif dan Gram negatif, mengetahui nilai konsentrasi hambat minimum (KHM) ekstrak daun ashitaba terhadap bakteri patogen Gram positif dan Gram negatif. Penelitian bersifat eksperimen, menggunakan Rancangan Acak Lengkap, analisis data dilakukan dengan uji One way anova dan uji lanjut Tukey. Perlakuan menggunakan 6 konsentrasi ekstrak etanol 70% daun ashitaba (0,10; 0,08; 0,06; 0,04; 0,02 dan 0,01) g/mL diujikan terhadap bakteri patogen Gram positif yaitu Methicilin Resisten Staphylococcus aureus (MRSA) dan bakteri patogen Gram negatif Pseudomonas aeruginosa diukur diameter rata-rata zona hambat (mm) dengan ulangan sebanyak 4 kali. Hasil penelitian menunjukan ekstrak etanol daun ashitaba berpengaruh signifikan (p<0,05) dalam menghambat aktivitas pertumbuhan bakteri MRSA dan P. aeruginosa, terdapat perbedaan diameter rata-rata zona hambat diantara variasi konsentrasi ekstrak etanol daun ashitaba pada bakteri MRSA dan P. aeruginosa. Nilai KHM bakteri MRSA adalah 0,10 g/mL dengan diameter rata-rata zona hambat sebesar 11,22 mm dan P. aeruginosa adalah 0,04 g/mL dengan diameter rata-rata zona hambat sebesar 8,04 mm. |
| Abtrak (Bhs. Inggris) | Ashitaba is medicinal plant, it is originated from Japan, in Indonesia unknown is potential biology. The previous research results suggested that plants contain secondary metabolites with antibacterial such as alkaloid, flavonoid, tanin, saponin, that are antimicrobial. Research of antibacterial activity test needs to determine the antibacterial activity potential of ethanol extract of ashitaba leaves that grow in Indonesia. The aim of this reserach are 1) to know the effect of ashitaba leaves 70% ethanol extract in inhibiting pathogenic bacteria Gram (+) and pathogenic bacteria Gram (-), 2) to know efect in inibiting MRSA was isolated from diabetic ulcers 3) to know the difference of antibacterial activity in each variation of the concentration extract and 4) to know the minimum inhibitory concentration (MIC) of ethanol extract of ashitaba leaves in inhibiting the growth of pathogenic bacteria Gram (+) and pathogenic bacteria Gram (-) in vitro. This research is an experimental design using completely random design (CRD). Antibacterial activity test carried out on six different treatments of concentration extract using diffusion method (Kirby Bauer). Preliminary test used concentration 0.1-1.0 g/mL. The next step is to the determine minimum inhibitory concentration by decrease the concentration 0.10; 0.08; 0.06; 0.04; 0.02, and 0.01 g/mL or increasing the concentration became 1.00; 2.00; 3.00; 4.00, and 5.00 g/mL. Antibacterial activity was analyzed through the test of honestly significant difference (HSD) using SPSS 18. The results of the research showed ashitaba leaf ethanol extract treatment concentrations 0.1-1.0 g/mL have antibacterial activity in inhibiting pathogenic bacteria Gram (+) Staphylococcus aureus, MRSA and Gram (-) Pseudomonas aeruginosa but not showed antibacterial activity to the bacteria Gram (-) Salmonella typhi. The treatment concentration extracts 0.10-0.01 g/mL significant effect (p<0.05) against S. aureus, MRSA, P. aeruginosa. The treatment concentration extracts 1.00-5.00 g/mL have significant effect (p<0.05) against the bacterium S. typhi. The results of HSD test showed the differences of antibacterial activity beetwen the treatment to bacteria pathogenic Gram (+) and bacteria pathogenic Gram (-). The minimum inhibitory concentration (MIC) of ashitaba leave ethanol extracts on pathogenic bacteria Gram (+) S. aureus and MRSA is 0.10 g/mL, pathogenic bacteria Gram (-) S. typhi is 2.00 g/mL and P. aeruginosa is 0.04 g/mL. The conclusion are ethanol extract 70% of ashitaba leaves that has infunced to inhibit the growth of pathogenic bacteria Gram (+) and pathogenic bacteria Gram (-). It this research showed the different avarage the inhibitor zone diameter on the treatment, MIC of pathogenic bacteria Gram (+) S. aureus and MRSA is 0.10 g/mL and pathogenic bacteria Gram (-) S. typhi is 2.00 g/mL, P aeruginosa is 0.04 g/mL. |
