| Abstrak (Bhs. Indonesia) | Latar Belakang: Paradoxical Sleep Deprivation (PSD) adalah metode induksi model stres kerja yang menyebabkan pengurangan waktu tidur tikus putih sehingga berefek pada kualitas reproduksi, yang ditandai oleh perubahan skor spermatogenik. Ekstrak etanol akar purwoceng mengandung eurikomalakton dan amarolinda, serta antioksidan yang bersifat pro spermatogenesis. Tujuan: Penelitian ini bertujuan untuk mengetahui perbedaan skor spermatogenik pasca pemberian ekstrak etanol akar purwoceng pada tikus putih jantan yang dipaparkan model stres PSD. Metode: Penelitian ini bersifat eksperimental dengan post-test only with control group design. Tikus putih dibagi secara acak menjadi 6 kelompok (masing-masing 5 ekor): kelompok kontrol negatif (I), kelompok kontrol PSD tanpa sleep recovery (KII) dan dengan sleep recovery (KIII), serta kelompok PSD yang diberi ekstrak etanol akar purwoceng dengan dosis berturut-turut 16,65 mg; 33,30 mg; dan 50,25 mg/200 gramBB/hari (KIV, KV, dan KVI). Paparan model stres PSD dilakukan selama 96 jam, dilanjutkan dengan pemberian sleep recovery dan perlakuan pada 7 hari setelahnya. Skor spermatogenik diukur menggunakan skor Johnsen. Hasil: Uji Kruskal-Wallis menunjukkan perbedaan bermakna (p <0,05). Uji Mann-Whitney menunjukkan perbedaan bermakna (p <0,05) rerata skor spermatogenik antara KII dan KIII dengan KI dan/atau KIV. KI memiliki rerata skor spermatogenik tertinggi (8,80 ± 0,54), KII dan KIII memiliki skor terendah dengan nilai yang sama (7,20 ± 0,45), KIV 8,20 ± 0,45, KV 8,00 ± 0,71, dan KVI 7,80 ± 0,84. Kesimpulan: Terdapat perbedaan skor spermatogenik yang bermakna pasca pemberian ekstrak etanol akar purwoceng pada kelompok tikus putih yang dipaparkan model stres PSD. Inggris) | Background: Paradoxical Sleep Deprivation (PSD) is an effective method to induce modality of stress due to work exposures that cause sleep deprivation in which affect quality of reproduction of rats, characterized by alterations of spermatogenic score. Ethanol extract of purwoceng roots contains euricomalacton, amarolinda, and antioxidant that support spermatogenesis. Objective: The aim of the study was to know the difference of spermatogenic score after administration of ethanol extract of purwoceng roots in male white rats exposed by PSD. Methods: This research was an experimental using post-test only with control group design. Rats were randomly devided into 6 groups (5 each): negative control group (I), PSD control group without sleep recovery (II) and also with sleep recovery (III), groups of rats with PSD given ethanol extract of purwoceng roots 16,65 mg; 33,30 mg; and 50,25 mg/200 gramsBW/day respectively (KIV, KV, and KVI). Ninety six hours of PSD was exposed, followed by 7 days of sleep recovery and treatments. The spermatogenic score was measured by Johnsen’s score. Result: Kruskal-Wallis test showed significant differences (p <0,05). Mann-Whitney test showed significant differences (p <0,05) among KII and KIII with KI and/or KIV. KI had the highest mean score of spermatogenic (8,80±0,54) while KII and KIII had same score as the lowest (7,20 ± 0,45), KIV 8,20 ± 0,45, KV 8,00 ± 0,71, and KVI 7,80 ± 0,84. Conclusion: There were significant differences on spermatogenic score after administration of ethanol extract of purwoceng roots in groups of rats that had been exposed by PSD.
