PENGUJIAN BEBERAPA ISOLAT BAKTERI Pseudomonas fluorescens TERHADAP LARVA ULAT GRAYAK ( Spodoptera litura )

Penelitian ini bertujuan untuk : 1) Mengetahui patogenisitas bakteri Pseudo-monas fluorescens untuk mematikan larva Spodoptera litura, (2) Mengetahui instar larva Spodoptera litura yang rentan terhadap perlakuan bakteri P. fluorescens, (3)Memperoleh hasil isolat bakteri yang efektif untuk ulat S. litura. Penelitian dilaksanakan di Laboratorium Perlindungan Tanaman Fakultas Pertanian Universitas Jenderal Soedirman Purwokerto. Penelitian dilaksanakan selama 7 bulan, dari bulan Maret hingga Oktober 2013. Rancangan percobaan yang digunakan Rancangan Acak Kelompok faktorial. Faktor pertama terdiri dari enam taraf (bakteri P. fluorescens P8, P. fluorescens P16, P. fluorescens P60, bakteri ulat (BESE), Insektisida kimia (profenofos), dan kontrol) dan faktor kedua adalah instar larva (instar 2, 3, dan 4). Percobaan diulang 3 kali. Percobaan kedua adalah pengaruh aplikasi P. fluorescens terhadap peletakan dan fertilitas telur. Perlakuan terdiri dari enam perlakuan termasuk kontrol (bakteri P. fluorescens P8, P. fluorescens P16, P. fluorescens P60, bakteri ulat (BESE), insektisida kimia (profenofos), dan kontrol. Hasil penelitian menunjukkan bahwa isolat bakteri P. fluorescens P8, P. fluorescens P16 , P. fluorescens P60, bakteri ulat (BESE), mampu menyebabkan mortalitas pada larva sebesar 63,3, 66,7, 71,2, dan 72,2 persen Bakteri P. fluorescens tidak dapat menekan peletakan telur. Walaupun demikian, bakteri tersebut mampu menekan fertilitas telur.

The goal of this research was to: 1) Assess the pathogenicity of the bacterium Pseudomonas fluorescens on larvae of Spodoptera litura, (2) Determine instar larvae of S. litura were susceptible to the bacterium Pseudomonas fluorescens treatment, (3) Obtain effective results for bacterial isolates S. litura. The experiment was conducted at the Laboratory of Plant Protection, Faculty of Agriculture, General Soedirman University, Purwokerto. The experiment was conducted for 7 months, from March to October 2013. Experimental design used a factorial randomized block design. The first factor consisted of six level (the bacteria P. fluorescens P8, P. fluorescens P16, P. fluorescens P60, (BESE), chemical insecticide (profenofos), and control) and the second factor was instar larvae (instar 2, 3, and 4) . The experiment was repeated 3 times. The second experiment was effect of P. fluorescens application on egg laying and fertility. Consisted of five treatments including the control (bacteria P. fluorescens P8, P. fluorescens P16, P. fluorescens P60, (BESE), chemical nsecticides (profenofos), and control. Results showed P. fluorescens isolate P8, P16 P. fluorescens, P. fluorescens P60, bacterial caterpillar (BESE), capable of causing mortality of larvae was 63.3, 66.7, 71.2, and 72.2 percent. P. fluorescens can not suppress egg laying. However, the bacteria was able to suppress the fertility of eggs.
