Optimasi Patch Transdermal Captopril Dengan Enhancer Asam Oleat, Plasticizer Propilen Glikol dan Cosolvent Isopropil Alkohol

Captopril merupakan obat golongan Angiotensin Converting Enzyme Inhibitor. Waktu paruh eliminasi captopril berkisar 1,6-1,9 jam, adanya makanan mengurangi absorbsinya menjadi 30-50%. Tujuan penelitian ini adalah untuk mengetahui formula optimum dan karakteristik fisik patch transdermal captopril menggunakan propilen glikol, asam oleat dan isopropil alkohol dalam polimer HPMC dan eudragit RL100 yang diharapkan dapat menjadi alternatif bentuk sediaan captopril. Formula patch transdermal captopril dirancang menggunakan simplex lattice design. Tipe patch transdermal adalah drug matrix in adhesive dengan memvariasikan propilen glikol, asam oleat, dan isopropil alkohol. Evaluasi karakteristik fisikkokimia patch transdermal meliputi visual, ketebalan, bobot, susut pengeringan, daya serap kelembaban, ketahanan terhadap pelipatan dan drug content formula optimum. Data yang diperoleh dari evaluasi sifat fisik dianalisis secara deskriptif menggunakan software design expert versi 7. Sedangkan verifikasi formula optimum menggunakan software open state versi 2012. Karakteristik fisik patch transdermal captopril dengan enhancer asam oleat, plasticizer propilen glikol dan cosolvent isopropil alkohol menghasilkan ketebalan 0,30 mm-0,60 mm, bobot 113 mg-161,33 mg, susut pengeringan 2,43%-14,24%, daya serap kelembaban 2,28%-16,42% dan ketahanan terhadap pelipatan 122 kali- 300 kali. Komposisi formula optimum yaitu 5% propilen glikol, 3% asam oleat dan 8% isopropil alkohol dengan ketebalan 0,29 mm, bobot 123 mg, susut pengeringan 4,04 %, daya serap kelembaban 2,96 %, ketahanan terhadap pelipatan 300 dan kandungan zat aktif 101,90%.

Captopril is a drug class Angiotensin Converting Enzyme Inhibitors. Elimination half-life of captopril range from 1,6-1,9 hours, the presence of food reduces the absorbs becomes 30-50%. The purpose of this study is to determine the optimum formula and physical characteristics of captopril patches using propylene glycol, oleic acid and isopropyl alcohol in the polymer HPMC and eudragit RL100 is expected to be an alternative dosage forms of captopril. Formula captopril transdermal patch designed using simplex lattice design. Type of transdermal patch was a drug in adhesive matrix with varying propylene glycol, oleic acid and isopropyl alcohol. Characteristic of evaluation of the physicochemical of transdermal patches included visual, thickness, weight, loss on drying, moisture uptake and folding endurance and drug content optimum formula. The data was obtained from the evaluation of the physical properties were analyzed descriptively using software Design Expert version 7. While the optimum formula was verified using software Open State version 2012. The physical characteristics of captopril transdermal patch with oleic acid enhancers, propylene glycol plasticizer and isopropyl alcohol cosolvent produces the thickness of 0,30 mm-0,60 mm, weight of 113 mg-161,33 mg, loss on drying of 2,43%-14,24%, moisture uptake of 2,28%-16,42% and folding endurance of 122-300. The proportion of the optimum formula is 5% of propylene glycol, 3% of oleic acid and 8% of isopropyl alcohol with a thickness of 0,29 mm, weight of 123 mg, loss on drying at 4,04%, moisture uptake at 2,96%, folding endurance of 300 and 101,90% of the drug content.
