Seiring berkembangnya teknologi saat ini menuntut kita untuk menciptakan inovasi baru di berbagai bidang, tidak terkecuali bidang medis seperti dalam hal  metode identifikasi sel darah. Analisis yang dilakukan oleh dokter berdasarkan preparat darah ini tidak selalu sama antara dokter yang satu dengan dokter yang lain. Ketelitian dan konsentrasi dokter sangat menentukan hasil analisis. Proses secara keseluruhannya sampai akhirnya diputuskan jenis penyakit yang diderita memakan waktu sekitar kurang lebih tiga hari. Sehingga diperlukan suatu sistem yang dapat melakukan identifikasi sel darah salah satunya berdasarkan morfologi  yaitu menghitung panjang diameter sel darah putih  menggunakan pengolahan citra digital. Sel darah tidak hanya mengandung sel darah putih, sehingga untuk mendapatkan bagian sel darah putihnya saja perlu dilakukan pendeteksian salah satunya menggunakan metode template matching. Selain itu untuk mendapatkan morfologi sel darah putih yang optimal diperlukan operasi morfologi untuk menghilangkan  noise – noise di sekeliling sel darah putih. Diameter merupakan adalah jarak terpanjang antara dua titik dalam tepi objek. Sehingga untuk menghitungnya harus mencari kontur ekternal dari citra tersebut terlebih dahulu.

Along with the expansion of the current technology requires us to create new innovations in various fields, such as the medical field is no exception in terms of identification methods of blood cells. The doctor analysis based on blood preparations which  this is not always gets the same results among doctors that one with another doctor. The doctor’s precision and concentration strongly determine the results of the analysis. The process as a whole until finally decided what kind of illnesses suffered takes about approximately three days. So it needed a system which can make identification of blood cells, one based on morphology to calculate the length of the diameter of the white blood cells using digital image processing. Blood cell not only contains white blood cells so to get the leukocyte only, used detection method such as using template matching and morphological  operation to get the optimal leukocyte morphology. Because morphological operation is used to eliminate  the noisesaround the leukocyte. Diameter is the longest is the distance between any two points within the object. So to calculate it should search for the image of the ekternal contour in advance.
