KAJIAN ANTIOKSIDAN MINUMAN RUMPUT LAUT Sargassum crassifolium-JAVA TEA CELUP PADA PLASMA DARAH TIKUS HIPERKOLESTEROLEMIK

Minuman rumput laut Sargassum crassifolium-java tea celup merupakan minuman berbahan dasar rumput laut Sargassum crassifolium dan daun kumis kucing (Orthosiphon aristatus (BL.) Miq). Minuman tersebut mempunyai kadar dan aktivitas antioksidan yang tinggi sehingga diduga dapat menghambat reaksi oksidasi komponen struktur tubuh. Tujuan dari penelitian ini untuk membandingkan serta mengetahui nilai dan hubungan antara kadar dan aktivitas antioksidan pada plasma darah tikus hiperkolesterolemik yang diberikan minuman rumput laut Sargassum crassifolium-java tea celup dengan berbagai flavoring dan minuman tradisional berbahan dasar Sargassum dari Cilacap. Penelitian ini berbentuk eksperimental dengan menggunakan Rancangan Acak Lengkap (RAL) yang terdiri dari 6 perlakuan, yaitu: P1 = kontrol negatif (pakan standar), P2 = kontrol positif (pakan kolesterol), P3 = minuman Sargassum crassifolium-java tea celup dengan flavoring lemon, P4 = minuman Sargassum crassifolium-java tea celup dengan flavoring lemon + jahe, P5 = minuman Sargassum crassifolium-java tea celup dengan flavoring lemon + pekak, dan P6 = minuman tradisional berbahan dasar Sargassum dari Cilacap. Perlakuan tersebut dilakukan pengulangan sebanyak 7 kali, sehingga diperoleh 42 unit percobaan. Variabel yang dikaji meliputi kadar dan aktivitas antioksidan plasma. Hasil penelitian menunjukkan, semua perlakuan pemberian minuman dapat menghambat pembentukkan MDA plasma, minuman Sargassum crassifolium-java tea celup flavoring lemon + jahe dapat meningkatkan kadar total fenol plasma tetapi belum mampu meningkatkan kadar glutation tereduksi. Kesimpulan dari penelitian ini yaitu minuman Sargassum crassifolium-java tea celup dengan penambahan flavoring lemon + jahe mampu meningkatkan kadar dan aktivitas antioksidan plasma darah tikus hiperkolesterolemik.

Sargassum crassifolium seaweed-java teabag is a functional drink which made from Sargassum crassifolium’s seaweed and leaf of cat whiskers (Orthosiphon aristatus (BL.) Miq). It has a high antioxidant content and activity which expected to be able to reduce oxidation reaction in our body. The aims of this research was to study and to compare the value and correlation between antioxidant content and activity in plasma of hyper-cholesterol rats which had been given Sargassum crassifolium seaweed-java teabag with various flavoring and traditional drink obtained from Cilacap regency which made from Sargassum. Complete Randomized Design (RAL) factorial were used in the current experiment with six treatments, i.e negative control rat was given standard diet (P1), positive control rat was given hyper-cholesterol diet (P2), Sargassum crassifolium seaweed-java teabag with lemon flavoring and hyper-cholesterol diet (P3), Sargassum crassifolium seaweed-java teabag with lemon + ginger flavoring and hyper-cholesterol diet (P4), Sargassum crassifolium seaweed-java teabag with lemon + star anise flavoring and hyper-cholesterol diet (P5), and traditional drink from Cilacap which made from Sargassum (P6). Each treatment was replicated seven times therefore, there were 42 experimental units. The tested variables were antioxidant content and activity on plasma. The result of this research showed that every treatments which given both Sargassum crassifolium-java teabag and traditional drink was able to decrease establishment of plasma malonaldehyde. Sargassum crassifolium-java teabag with lemon + ginger flavoring was able to increase total phenolic content of plasma. However, it was not able to increase glutathione content of plasma. The conclusion of this research showed that Sargassum crassifolium-java teabag with lemon + ginger flavoring was able to increase antioxidant content and activity of hyper-cholesterol rats’ plasma.
