IDENTIFIKASI DAN VIRULENSI Ganoderma sp. ASAL KELAPA SAWIT DARI BEBERAPA WILAYAH DI INDONESIA

Kelapa sawit (Elaeis guineensis Jacq.) merupakan komoditas strategis Indonesia sebagai produsen minyak sawit terbesar dunia, namun produktivitasnya terancam oleh penyakit busuk pangkal batang (Basal Stem Rot) yang disebabkan oleh jamur patogen genus Ganoderma, terutama Ganoderma boninense. Penyakit ini bersifat "silent killer" karena gejala baru tampak setelah infeksi lanjut, menyebabkan kerugian produksi hingga lebih dari 50% pada lahan terinfeksi parah. Penelitian ini bertujuan mengidentifikasi spesies Ganoderma sp. dari berbagai wilayah di Indonesia, mengetahui tingkat virulensi setiap isolat, dan menentukan isolat dengan virulensi tertinggi. Penelitian dilaksanakan di Laboratorium Perlindungan Tanaman, Laboratorium Riset, dan Experimental Farm Universitas Jenderal Soedirman, Purwokerto, menggunakan delapan isolat Ganoderma sp. asal Papua, Sulawesi, Sumatera Utara, Jawa Barat, Jambi, Lampung, Riau, dan Indragiri Hilir Riau. Metode meliputi karakterisasi morfologi, uji aktivitas enzim lignolitik (lakase, mangan peroksida, lignin peroksida), identifikasi molekuler berbasis PCR dengan primer ITS1 dan ITS4, analisis BLAST dan pohon filogenetik, serta uji virulensi in planta menggunakan Rancangan Acak Kelompok (RAK) yang dianalisis dengan ANOVA dan DMRT (α = 5%). Hasil menunjukkan keragaman aktivitas enzim lignolitik antar isolat, dengan isolat IH dan GB- menunjukkan aktivitas enzim tertinggi. Analisis BLAST mengidentifikasi isolat GB-, IH, SL, dan JB sebagai G. boninense, sedangkan GB+, SSU, GJ, dan Papua sebagai G. australe. Uji virulensi in planta menunjukkan intensitas penyakit 34,38–53,13%, dengan isolat IH, SL, GB-, dan SSU (berkerabat G. boninense) menunjukkan virulensi tertinggi, sementara isolat berkerabat G. australe menunjukkan virulensi lebih rendah. Tinggi tanaman tidak berbeda nyata antar perlakuan.

Oil palm (Elaeis guineensis Jacq.) is a strategic commodity for Indonesia as the world's largest palm oil producer, but its productivity is threatened by basal stem rot disease caused by pathogenic fungi of the genus Ganoderma, particularly Ganoderma boninense. This disease is known as a "silent killer" because symptoms only appear after advanced infection, causing production losses of more than 50% in severely infected areas. This study aimed to identify Ganoderma sp. species from various regions in Indonesia, determine the virulence level of each isolate, and identify the most virulent isolate. The research was conducted at the Plant Protection Laboratory, Research Laboratory, and Experimental Farm of Universitas Jenderal Soedirman, Purwokerto, using eight Ganoderma sp. isolates from Papua, Sulawesi, North Sumatra, West Java, Jambi, Lampung, Riau, and Indragiri Hilir Riau. Methods included morphological characterization, lignolytic enzyme activity assays (laccase, manganese peroxidase, lignin peroxidase), molecular identification based on PCR using ITS1 and ITS4 primers, BLAST analysis and phylogenetic tree construction, and in planta virulence testing using a Randomized Block Design (RBD) analyzed by ANOVA and DMRT (α = 5%). Results showed variation in lignolytic enzyme activity among isolates, with isolates IH and GB- showing the highest absorbance. BLAST analysis identified isolates GB-, IH, SL, and JB as G. boninense, while GB+, SSU, GJ, and Papua were identified as G. australe. In planta virulence testing showed disease intensity ranging from 34.38–53.13%, with isolates IH, SL, GB-, and SSU (closely related to G. boninense) exhibiting the highest virulence, while isolates related to G. australe showed lower virulence. Plant height did not differ significantly among treatments.
