Biosintesis Nanopartikel Emas (AuNPs) Menggunakan Ekstrak Rimpang Kencur Hitam (Kaempferia parviflora) dan Uji Antioksidannya

Radikal bebas merupakan penyebab utama berbagai penyakit degeneratif sehingga diperlukan penawar seperti antioksidan yang tentunya efektif dan ramah lingkungan. Nanopartikel emas (AuNPs) berpotensi sebagai antioksidan karena aktivitasnya yang tinggi dan dapat disintesis secara biosintesis dengan memanfaatkan senyawa flavonoid dan polifenol dari ekstrak rimpang K. parviflora sebagai agen pereduksi sekaligus penstabil. Penelitian ini bertujuan mensintesis nanopartikel emas dari ekstrak air rimpang K. parviflora (KP-AuNPs) dengan variasi konsentrasi HAuCl4 1; 2,5; dan 5 mM, mengkarakterisasi KP-AuNPs menggunakan spektrofotometer UV-Vis, FTIR, PSA, dan SEM-EDX+Mapping, serta menguji aktivitas antioksidannya dengan DPPH dan ABTS. Hasil penelitian menunjukkan biosintesis KP-AuNPs ditandai perubahan warna larutan menjadi merah anggur dan puncak Surface Plasmon Resonance (SPR) pada 500–550 nm. Karakterisasi FTIR mengonfirmasi keterlibatan gugus hidroksil, karbonil dan eter, sedangkan PSA dan SEM-EDX secara berturut-turut menunjukkan ukuran partikel optimal pada konsentrasi 1 mM 42 nm dan 39,32 nm dengan distribusi unsur Au yang merata. Uji antioksidan DPPH menunjukkan KP-AuNPs konsentrasi 1 mM memiliki aktivitas tertinggi dengan nilai IC50 3,83 mM, sedangkan pada ABTS memiliki IC50 3,49 mM. Hasil kedua metode DPPH dan ABTS memiliki nilai yang lebih baik dibandingkan ekstrak tanpa perlakuan. Ekstrak K. parviflora efektif untuk biosintesis AuNPs dengan potensi aktivitas antioksidan yang dimiliki.

Free radicals are major contributors to various degenerative diseases, highlighting the need for effective and environmentally friendly countermeasures such as antioxidants. Gold nanoparticles (AuNPs) have potential as antioxidant agents owing to their high activity and can be synthesized through a green biosynthetic approach using flavonoids and polyphenols present in Kaempferia parviflora rhizome extract as both reducing and stabilizing agents. This study aimed to synthesize gold nanoparticles using an aqueous extract of K. parviflora rhizomes (KP-AuNPs) at varying HAuCl4 concentrations of 1, 2.5, and 5 mM; characterize the resulting KP-AuNPs using UV–Vis spectrophotometry, FTIR, PSA, and SEM–EDX+mapping; and evaluate their antioxidant activity using DPPH and ABTS assays. The results showed that KP-AuNP biosynthesis was indicated by a change in solution color to wine red and the appearance of a surface plasmon resonance peak at 500–550 nm. FTIR characterization confirmed the involvement of hydroxyl, carbonyl, and ether functional groups, while PSA and SEM–EDX analyses showed that the 1 mM formulation yielded optimal particle sizes of 42 nm and 39.32 nm, respectively, with a homogeneous distribution of Au. The DPPH assay showed that KP-AuNPs synthesized at 1 mM exhibited the highest antioxidant activity, with an IC50 value of 3.83 mM, whereas the ABTS assay yielded an IC50 value of 3.49 mM. The antioxidant activities determined using both DPPH and ABTS assays were superior to those of the untreated extract. These findings demonstrate that K. parviflora extract is effective for the green biosynthesis of AuNPs and that the resulting nanoparticles exhibit promising antioxidant potential.
