Akumulasi Prolin pada Daun Mangrove di Muara Sungai Pemali, Kabupaten Brebes

Ekosistem mangrove di Muara Sungai Pemali, Kabupaten Brebes, rentan terhadap perubahan kualitas lingkungan yang dapat memicu stres fisiologis pada tanaman. Penelitian ini bertujuan menganalisis kadar prolin pada daun mangrove, kondisi kualitas air, serta hubungan antara kualitas air dan akumulasi prolin. Penelitian dilaksanakan pada Agustus--September 2025 di tiga stasiun dengan perbedaan umur tegakan mangrove (17, 7, dan 3 tahun). Parameter yang diukur meliputi suhu, pH, DO, TDS, salinitas, nitrat, fosfat, dan kadar prolin. Data dianalisis menggunakan ANOVA, Kruskal-Wallis, korelasi Pearson, dan regresi linier berganda. Hasil penelitian menunjukkan kadar prolin tertinggi pada Acanthus ilicifolius (1,143 mg/g BB) dan Avicennia marina (0,855 mg/g BB), serta terendah pada Rhizophora mucronata (0,229 mg/g BB), namun secara statistik tidak berbeda nyata (p>0,05). Parameter kualitas air (suhu, pH, DO, TDS, salinitas, nitrat) berbeda nyata antarstasiun (p<0,05), kecuali fosfat. Analisis regresi menunjukkan suhu (β=0,893) dan DO (β=-0,432) merupakan parameter paling berpengaruh terhadap akumulasi prolin. Akumulasi prolin mencerminkan respons adaptif mangrove terhadap tekanan lingkungan, dengan suhu dan DO sebagai indikator paling representatif dalam menggambarkan tingkat stres fisiologis mangrove di Muara Sungai Pemali.

Mangrove ecosystems in the Pemali River Estuary, Brebes Regency, are vulnerable toward the environmental change that can trigger physiological stress in plants. This study aimed to analyze proline content in mangrove leaves, water quality conditions, and the relationship between water quality and proline accumulation. The study was conducted from August to September 2025 at three stations representing different mangrove stand ages (17, 7, and 3 years). Parameters measured included temperature, pH, DO, TDS, salinity, nitrate, phosphate, and proline content. Data were analyzed using ANOVA, Kruskal-Wallis, Pearson correlation, and multiple linear regression. The results showed the highest proline content in Acanthus ilicifolius (1.143 mg/g WW) and Avicennia marina (0.855 mg/g WW), and the lowest in Rhizophora mucronata (0.229 mg/g WW), but statistically not significantly different (p>0.05). Water quality parameters (temperature, pH, DO, TDS, salinity, nitrate) differed significantly among stations (p<0.05), except phosphate. Regression analysis showed temperature (β=0.893) and DO (β=-0.432) as the most influential parameters on proline accumulation. Proline accumulation reflects the adaptive response of mangroves to environmental stress, with temperature and DO being the most representative indicators of physiological stress levels in mangroves at the Pemali River Estuary.
