IDENTIFIKASI MOLEKULER DAN PROFIL SENYAWA BIOAKTIF RUMPUT LAUT Halymenia sp. DARI TELUK EKAS, NUSA TENGGARA BARAT

Halymenia sp. merupakan rumput laut merah yang memiliki potensi senyawa bioaktif dan berpotensial diterapkan dalam berbagai industri. Penelitian ini bertujuan untuk mengidentifikasi secara molekuler dan menunjukkan profil senyawa bioaktif Halymenia sp. dari Teluk Ekas, Nusa Tenggara Barat. Identifikasi molekuler Halymenia sp. menggunakan metode DNA Barcoding gen ribulose-1,5-biphosphate carboxylase/oxygenase large subunit (rbcL) dan sanger sequencing. Profil senyawa bioaktif diperoleh dari metode ekstraksi maserasi bertingkat dan MAE serta analisis metabolomik berbasis liquid chromatography high resolution mass spectrometry (LC-HRMS) dan pendekatan jaringan molekuler. Seluruh spektrum spektrometri massa dari ekstrak Halymenia sp. dikonversi menjadi file .mzXML menggunakan MSConvert, diunggah ke Global Natural Products Social (GNPS), dan divisualisasi menggunakan Cytoscape 3.9.1. Berdasarkan hasil penelitian, identifikasi morfologi dan anatomi serta identifikasi molekuler memiliki hasil yang selaras, yakni memiliki hubungan kekerabatan dengan Halymenia durvillei sebesar 98,38%. Profil senyawa bioaktif yang diperoleh antara lain cholecalciferol (m/z 385,376 [M+H]), 7-dehydrocholesterol (m/z 387,322 [M+H]), pheophorbide A (m/z 593,269 [M+H]), dan pyropheophorbide A (m/z 535,263 [M+H]). Kesimpulannya, melalui keberhasilan identifikasi spesies dan senyawa bioaktif ini, diharapkan dapat diaplikasikan dalam pemanfaatan terapan atau penelitian lanjutan untuk mendapatkan hasil yang lebih akurat.

Halymenia sp. is a red seaweed that has potentials for bioactive compounds and potential to be applied in various industries. This study aimed to molecularly identify and profile the bioactive compounds of Halymenia sp. from Ekas Bay, West Nusa Tenggara. Molecular identification used DNA barcoding of ribulose-1,5-biphosphate carboxylase/oxygenase large subunit (rbcL) and sanger sequencing. Meanwhile, bioactive profiling was performed using sequential maceration, microwave-assisted extraction (MAE), liquid chromatography high resolution mass spectrometry (LC-HRMS)-based on metabolomics, and molecular networking. Raw mass spectrometry data were converted to .mzXML files via MSConvert, processed on the GNPS platform, and visualized using Cytoscape 3.9.1. Based on the results of the study, morphological and anatomical identification and molecular identification have consistent results, namely having a similarity with Halymenia durvillei of 98.38%. The metabolomic profiles obtained included cholecalciferol (m/z 385,376 [M+H]), 7-dehydrocholesterol (m/z 387,322 [M+H]), pheophorbide A (m/z 593,269 [M+H]), and pyropheophorbide A (m/z 535,263 [M+H. In conclusion, through the successful identification of these bioactive species and compounds, it is hoped that they can be applied or further research to obtain more accurate results.
